Summary Immunological mechanisms are demonstrably of central importance in preventing the development of certain virus‐associated cancers in animals and man; however, they do not appear to fulfil this role in the majority of ‘spontaneous’ tumours. This does not, however, necessarily indicate that spontaneous tumours lack potential target antigens for immunologically mediated destruction. Work in the field of transplantation immunology has clearly shown that certain cell types fail to elicit rejection reactions despite their possession of alloantigens. Similarly, some tumour cell types are poorly immunogenic to the point that they can grow in and kill animals despite a major histocompatability barrier. These tumours are, however, susceptible to destruction in vivo in appropriately allo‐sensitized animals. Thus, some tumours may be able to grow in autologous or syngeneic hosts because of their poor immunogenicity, despite the fact that they express potential (tumour‐associated) rejection antigens. It may be possible to manipulate this situation for therapeutic purposes.

Previous reports indicate that human hepatocytes do not express class I and class II MHC antigens. Our analyses on 10 human hepatocellular carcinoma (HCC) cell lines by immunofluorescence tests and RIA, demonstrate that all the human HCC cell lines tested express class I MHC antigens and among them, three poorly differentiated human HCC cell lines also express class II MHC antigens. Results of immunoprecipitation and/or Western blotting experiments indicate similarity in the chemical nature of both the class I and class II MHC antigens expressed by the human HCC cell lines and by a human B lymphoblastoid cell line Raji. Furthermore, a new variant form of class I antigen was detected in some of these HCC cell lines. Immunohistochemical studies of HCC tissues using the peroxidase-antiperoxidase staining method indicated that class I and class II antigens were detectable in 7 out of 11 and 3 out of 11 HCC tissues from patients, respectively. The availability of MHC class I antigen-positive cultured HCC cell lines, including the poorly differentiated lines that also express MHC class II antigen, has provided us with interesting models to study the relationship between expression of MHC antigen and transformation and differentiation of human hepatocytes. These studies will also allow us some insight into the role of MHC class I and class II antigen in the immunosensitivity and immunogenecity of HCC cells to the host-immune response.

Expression of class I MHC transplantation antigens has been shown to be reduced in baby rat kidney (BRK) cells transformed by highly oncogenic adenovirus type 12 (Ad12), as compared with untransformed cells and cells transformed by non‐oncogenic Ad5. Here we show that this reduction of class I expression also occurs in a variety of other primary cell cultures transformed by Ad12, and that reduction of class I gene expression occurs for all class I loci. Transfection of Ad5E1 into class I‐negative Ad12‐transformed BRK cells leads to complete restoration of class I expression. Introduction of Ad12E1 into most class I‐positive established cell lines does not result in suppression of class I expression. However, transfection of the Ad12E1A region into a class I‐positive cell line which was immortalized by a mutant Ad12E1A region resulted in suppression of class I gene expression, implying that the suppression of class I activity in Ad12‐transformed cells is due to an active switching‐off process.
