Variations in inflammation-related genes may be associated with childhood febrile seizure susceptibility

Abstract: Genetic variants in inflammation-related genes, specifically purinergic receptor P2X7, may be involved in susceptibility to childhood-onset febrile seizures.

“…Khoshdel et al found a significant correlation between genotype distribution of G-308A and complex FS in Iranian patients, but there was no correlation between TNF-α polymorphisms at positions of −308A, and 850T and predisposition to simple FS [4]. Emsley et al found no significant association of TNF-α SNPs, but significant result was for missense SNP rs208294 in P2X7R (purinergic receptor P2X7) [41]. Also, Chou et al demonstrated that IL-1 receptor antagonist allele 1 homozygote was more frequent in patients with FS than in healthy controls, but no significant difference was evident in the distribution of allele frequencies of the IL-1B promoter, IL-1B exon 5, IL-6 promoter, IL-8, IL-10, or the TNF-α gene polymorphism [42].…”

“…Some studies showed that MPVs levels were increased due to decreased platelet count during acute infection. Therefore platelets play an important role during infection and inflammation 28,29 . Some other studies suggests that inflammation, which is intrinsic to the fever response, is involved in the generation of FS 15 .…”

“…Inflammatory response components, such as those involved in the interleukin 1 (IL-1) system, have been suspected to play a role in FS [Gatti et al, 2002]. Recent reports indicate that certain SNPs in genes that encode other inflammation-related molecules, such as IL-6 [Shahrokhi et al, 2014], IL-6 receptor, prostaglandin E receptor 3, purinergic receptor P2X7, or TLR-4, may be involved in the pathogenesis of FS [Emsley et al, 2014].…”
