Vasoactive agents affect growth and protein synthesis of cultured rat mesangial cells

Abstract: Mesangial cell (MC) proliferation and extracellular matrix (ECM) formation are hallmarks of chronic glomerular disease. The present in vitro study examined the effects of the vasoactive agents angiotensin II (Ang II), arginine vasopressin (AVP), and serotonin (5-HT) on growth and protein biosynthesis of cultured rat MCs after 72 hours of incubation. AVP and 5-HT (10(-6) M) significantly increased DNA synthesis and growth of quiescent subconfluent MCs to levels of 25 and 45%, respectively, of the optimal stimul… Show more

“…This observation agrees with other reports that also directly linked the rate ofcollagen synthesis with that ofmesangial and arterial smooth muscle cell growth in culture (63)(64)(65). The in vivo relevance of these findings is evidenced by the progressive glomerulosclerosis occurring in the hypertrophied glomeruli of mice transgenic for growth hormone, where excessive deposition of ECM is closely associated with increased cell turnover (66).…”

“…3-mo-old male Wistar-Kyoto rats (WKY) were fed a standard pellet diet and water ad libitum. According to previously described methods ( 17,(19)(20)(21) Precipitation on ice was performed with 200 yd 20% trichloroacetic acid (TCA) overnight. TCA-precipitable material was pelleted by centrifugation at 2,000 g, dissolved in 0.5 M NaOH, and transferred for liquid scintillation counting.…”

“…Since a number of vasopressor hormones, such as angiotensin II, vasopressin and endothelin, also act as growth factors on vascular smooth muscle cells or MC (17,18), it was tested in the present study, whether also AP5A and AP6A stimulate growth of MC. Furthermore, both dinucleotides were tested for interactions with polypeptide growth factors.…”

“…The supernatants were assayed for released fibronectin, while washed cell layers were sonicated in 500 /iL of 1% nonidet P-40 in PBS and assayed for cell-associated fibronectin. Fibronectin levels in all samples were determined by a double-antibody sandwich enzymelinked immunosorbent assay using the methods provided by the manufacturer (DAKO Corp, Carpinteria, Calif) and Wolthuis et al 12 A polyclonal rabbit anti-human fibronectin (1:1000) was used as the coating antibody, and the detection antibody was a peroxidase-conjugated rabbit anti-human fibronectin (1:2000).…”
