Alternative to xylene as a clearing agent in histopathology

INTRODUCTION: Clearing is an essential step in processing tissue for light microscopy. Xylene is the clearing agent used most commonly worldwide. Xylene is toxic and therefore a threat to personnel working in histopathology laboratories. We evaluated a safer alternative clearing agent for use in the histopathology laboratory. MATERIALS AND METHODS: We used 230 formalin-fixed, paraffin-embedded tissue blocks from 19 different tissues. Half of the specimens were processed using xylene and half were processed using UltraClear™. Tissues were evaluated for eight parameters: sectioning, nuclear staining, cytoplasmic staining, overall cell morphology, clarity of staining, uniformity of staining, quality of immunohistochemistry (IHC), and cost. RESULTS: Both UltraClear™ and xylene processed sections scored 100% for IHC. Sections processed using UltraClear™ were easy to cut (81.7%) as were xylene processed sections (96.5%). UltraClear™ processed sections showed 67%, 60.9%, 52.2%, 63.5%, and 67% for nuclear staining, cytoplasmic staining, cell morphology, clarity of staining, and uniformity of staining, respectively. UltraClear™ is twice as expensive as xylene. We found that tissues processed using UltraClear™ were easy to cut and worked well for both hematoxylin and eosin and IHC staining. CONCLUSION: UltraClear™ is less toxic, less flammable, friendlier to the environment, and easy to handle, but it is two times expensive than xylene. The findings of this study recommend the use of UltraClear™ solution as a routine clearing agent in histopathology laboratories. However, further studies are required.

Cell and sub‐cellular anatomical adjustments are adaptations utilized by plants to tolerate abiotic stress. Both melatonin and Morinda lucida‐silver nanoparticles (ML‐AgNPs) are recognized as bio‐stimulants. The study examined the morphological changes and adaptive characteristics of these bio‐stimulants under water‐stress Eugenia uniflora. Twenty‐four hours was spent priming the seeds with melatonin (0.06 mg/L), ML‐AgNPs (0.06 mg/L), and a mixture (1:1) of the two. The seeds were sown and subjected to water stress for 7 days. The leaves, stems, and roots of water‐stressed E. uniflora were sectioned, dried, and examined using a microscope. Drought stress led to the production of non‐glandular trichomes on the abaxial and the transformation of paracytic stomata into diacytic stomata. During water stress, melatonin enlarges intercellular gaps and stomata, increases sponge and palisade parenchyma, and thickens epidermis (stem and root) and fibers. The ML‐AgNPs diminished the size of mesophyll, intercellular gaps, stomata, and stem fiber. The ML‐AgNPs increased the size of bulliform cells and activated the mechanical resistance features of sclerophyllous leaves (thick‐celled epidermis and sclerieds) and ray parenchyma (root and stem). Equally, Melatonin and ML‐AgNPs increased stem and root anatomical characteristics (xylem, bark, pith, cortex, epidermis, and vascular bundles). Stomata of E. uniflora are susceptible to alterations and undergo cell division into two new stomata (stomatogensis) in response to varying conditions (melatonin and ML‐AgNPs). Melatonin adopted a strategy for maintaining a high plant water status, possibly by osmoregulation, whereas E. uniflora primed with ML‐AgNPs survived by minimizing transpirational water loss through morphological changes.

Clearing results in histological processing can be affected due to tissue density and viscosity of the clearing agent. xylene is the most commonly used aromatic solvent for clearing agents and deparaffinizing agents in histopathology laboratories, but xylene is one of the most dangerous chemicals found in histology laboratories. Based on the dangers posed by xylene, a replacement material is needed. Several xylene substitutes such as reagents derived from limonene, aliphatic hydrocarbons, vegetable oils and mineral oils have been commercially developed, but the available xylene substitutes are still less effective, more expensive, and still as dangerous as xylene itself. Substitutes that are considered safer are from natural oils, one of which is olive oil which has similarities with xylene, namely in hydrocarbon and phenol compounds. This study used two types of tissue, namely skin and liver from mice (Mus musculus) which were cut into two parts; the first part uses xylene as and the other part uses olive oil as a clearing agent. The assessment of hematoxylin eosin staining results were determined based on the category of assessment of cell structure, such as the cell nucleus, cytoplasm, and color uniformity carried out by three readers and five fields of view for each microscopic slide at 40X magnification of the objective lens. Comparison of observations of the xylene group in skin and liver tissue 100% got a good score on the cell nucleus, cytoplasm, and color uniformity. (p=1,000). The olive oil group had a slight difference in color uniformity in liver tissue when compared to skin, but not statistically different (p=0.773). The comparison of the overall readings of the xylene and olive oil groups on skin and liver tissue also showed no statistically different (p=0.262). So it can be concluded that olive oil can be recommended as a substitute for xylene in the clearing process in histological tissue processing.
