“…The membrane components which are mobilized during anesthetic treatment to form swellings are not similar to the structures referred to as 'mounds' [29] or blebs [12] which represent artifacts of glutaraldehyde fixation. The evidence for distinguishing mounds from the swellings observed in this study is as follows: (a) the swellings are easily visualized in the light microscope prior to fixation; (b) the swellings encompass the en tire fiber; (c) the swellings contain a wide variety of membrane components other than agranular vesicles, such as smooth endoplas mic reticulum, autophagic vacuoles, dense core bodies and coated vesicles; and (d) struc tures identical to mounds are occasionally seen in electron micrographs ( fig.9e) but are not located in the swollen regions.…”

“…Although vesicles and smooth endoplasmic reticulum are seen throughout the length of the neurite, they are often clustered in expansions along the periphery of the nerve fiber. Some of these swellings have the appearance of varicosities which have been observed in other neurons in culture [16,47], The bags of membraneous sacs, termed 'mounds' [29], are only occasion ally seen.…”

“…The appearance of such images, referred to as mounds, has been suggested to result from fixation procedures (Nuttall & Wessells, 1979). But even if this is the case they are only observed in developing tissues at certain restricted areas.…”

“…Most of our current understanding of growth-cone function has been elucidated by in vitro time-lapse studies [Bray et al, 1978;Letourneau, 1979;Nuttall and Wessells, 1979;Landis, 1983; Bray and Chapman, 19851. Studies of axon extension in vivo have demonstrated that these data are consistent with growthcone behaviour examined in the natural context [Raper et al, 1983;Mason, 1985;Harris et al, 1985; Tosney and Landmesser, 19851.…”
