VIRMA-Dependent N6-Methyladenosine Modifications Regulate the Expression of Long Non-Coding RNAs CCAT1 and CCAT2 in Prostate Cancer

Abstract: RNA methylation at position N6 in adenosine (m6A) and its associated methyltransferase complex (MTC) are involved in tumorigenesis. We aimed to explore m6A biological function for long non-coding RNAs (lncRNAs) in prostate cancer (PCa) and its clinical significance. m6A and MTC levels in PCa cells were characterized by ELISA and western blot. Putative m6A-regulated lncRNAs were identified and validated by lncRNA profiler qPCR array and bioinformatics analysis, followed by m6A/RNA co-immunoprecipitation. Impact… Show more

“…METTL3 promotes development, progression and metastasis of PRAD through regulating MYC methylation, hedgehog pathway, and Wnt pathway (Cai et al, 2019;Ma et al, 2020;Yuan et al, 2020) and enhances bone metastasis through m 6 A-HuR-dependent mechanism (Li et al, 2020). VIRMA sustains invasiveness of PC cell through regulating oncogenic lncRNAs (Barros-Silva et al, 2020). YTHDF2 promotes the proliferation and migration of PC that is suppressed by miR-493-3p (Li et al, 2018).…”

“…Besides, METTL3 and VIRMA were also reported promoting the development and progression of PRC in different manners. [150][151][152][153] Renal Cell Carcinoma. METTL3 and METTL14 can not only promote the proliferation and metastasis of tumor cells, but also inhibit the progress of tumor.…”

“…The knockout of VIRMA triggers a compensatory feedback loop that enhances the expression of the catalytic METTL3 subunit. However, compensatory METTL3 overexpression is insufficient to maintain MTC function without VIRMA (Barros-Silva et al, 2020) (Fig. 3).…”
