Virus Induction of Osteosarcomas in Mice

Abstract: A virus extractcd from an osteosarcoma of a mouse produces simiillar fumors when Injected into newborn mice. The original tumor appeared spoiltaneously in an untreated CFI male. The time between inoculation with virus and detection of bone sarcoma has been as short as 23 days.

“…2 The viral fos oncogene (v fos) causes a very specific osteogenic neoplasm in mice (Finkel et al 1966). It seems that v fos transforms a pre-bone cell that retains the capacity for both osteodi and chondroid differentiation.…”

“…The c-fos proto-oncogene is the cellular homolog of the transforming gene of FBJ osteosarcoma virus (6,9,12) and encodes a nuclear protein that is thought to be involved in normal cellular growth and differentiation (7,35). In a number of different cell types, transcription of c-fos is rapidly and transiently activated by growth factors (1,8,19,22,25,26,31,32).…”

“…Several methods to induce bone tumors were undertaken ; zink beryllium (12), radioactive compounds (11 , 19, 24, 48), chemical carcinogens (19,22,46), and X-ray irradiation (9,40) . Recently a role played by a virus in the oncogenic process of osteosarcoma has been reported (10). The original tumor of Gardner mouse osteosarcoma 519 was found in a CS 7BL~ x CBA Fl hybrid mouse which had received 175R of X-rays.…”
