Viruses from extreme thermal environments

Abstract: Viruses of extreme thermophiles are of great interest because they serve as model systems for understanding the biochemistry and molecular biology required for life at high temperatures. In this work, we report the discovery, isolation, and preliminary characterization of viruses and virus-like particles from extreme thermal acidic environments (70 -92°C, pH 1.0 -4.5) found in Yellowstone National Park. Six unique particle morphologies were found in Sulfolobus enrichment cultures. Three of the particle morphol… Show more

“…Enrichment cultures were established from a YNP acidic hot spring (pH 2.9-3.9, 72-92°C) in the Rabbit Creek thermal area (44°31.287ЈN, 110°48.647ЈW) within the Midway Geyser Basin, as described (4). A single-colony isolate of Sulfolobus solfataricus expressing a spherical virus-like particle was established by previously described methods (4).…”

“…Isolate YNPRC179 was inoculated into medium 88 (www.dsmz.de͞species͞gn301555.htm), and the culture supernatants were monitored for virus production by direct particle counts with a transmission electron microscope (TEM), as described (4). When virus titers reached maximum levels (typically 3 days), cells were removed by centrifugation (6,000 ϫ g for 10 min) and the supernatants filtered through Acrodisc PF 0.8͞0.2-m filters (Pall Gelman Laboratory, Ann Arbor, MI).…”

“…Electron microscopy observations suggest that linear viruses dominate in high-temperature environments (12,33,35,42). Those which have been isolated and characterized to date infect members of the archaeal genera Sulfolobus, Acidianus, and Thermoproteus.…”

“…These extreme environmental conditions favor not only the Archaea but also result in a relatively simplified microbial community structure. A number of viruses, exclusively archaeal, have been isolated out of these environments (Rice et al, 2001;Mochizuki et al, 2011;Pina et al, 2011;Prangishvili, 2013). These conditions offer a tractable system to study virus-host relationships, as well as viral and host community structure and stability.…”
