VisEgg: a robust phenotyping tool to assess rainbow trout egg features and viability

Abstract: HAL is a multi-disciplinary open access archive for the deposit and dissemination of scientific research documents, whether they are published or not. The documents may come from teaching and research institutions in France or abroad, or from public or private research centers. L'archive ouverte pluridisciplinaire HAL, est destinée au dépôt et à la diffusion de documents scientifiques de niveau recherche, publiés ou non, émanant des établissements d'enseignement et de recherche français ou étrangers, des labor… Show more

“…Inversely, when eggs are of bad quality, the vitelline membrane is weakened and the water pressure is enough to break it. In this case, eggs turns white during hydration [35]. The higher fry percent survival after resorption also confirmed higher egg quality from females fed the plant-based diet.…”

“…Stripped unfertilized eggs (approximatively 400 eggs, about 25 to 30 g) were placed in 150 mL containers and water was added to hydrate the eggs. After 24 h of hydration, a picture of the sampled eggs was obtained using the VisEGG shooting system consisting of a light tablet and digital SLR camera (canon EOS 1000D, resolution: 10.1 M pixels) equipment, as previously described [35]. Pictures were analyzed with Visilog 7.3 software (Thermo Scientific) allowing for automatic measurements to be made (number and size of the eggs and white egg percentage).…”

“…To do so, the eggs of each fertilization test were transferred into a petri dish and a picture was taken. The total number of eggs was counted with the macro VisEgg 59 on Fiji-ImageJ while the number of fertilized oocytes (embryos with developed eyes) was manually counted with the Fiji-ImageJ counter. Fertilization percentage was calculated by the following formula: (number of fertilized eggs/total number of eggs × 100).…”

“…Ovulation was monitored three time a week. On the control site, a total of 32 fish were used including 18 for egg quality assessment using the VisEgg system (Cardona et al 2020) (4 of them being also used for the oocyte hydration study and 11 of them being also used for blood sampling) and 14 for the molecular analysis. On the LSW site, a total of 46 fish were used including 30 for egg quality assessment using the VisEgg system (4 of them being also used for the oocyte hydration study and 9 of them being also used for blood sampling) and 16 for the molecular analysis.…”

“…Oocyte viability evaluation was systematically performed using the VisEgg protocol when ovulation occurred. This recently developed tool allows a robust assessment of different egg features including egg viability (Cardona et al, 2020). The process uses a standardized image-base batch analysis method of 24h-hydrated unfertilized eggs and an image processing algorithm.…”
