Visualising oxytocin neurone activity in vivo: The key to unlocking central regulation of parturition and lactation

Abstract: Although current research on oxytocin largely focusses on its behavioural effects, 1-3 oxytocin was originally identified as the trigger for uterine contraction during parturition and mammary duct contraction during suckling. 4 Although transgenic mice without oxytocin can give birth, 5 oxytocin receptor antagonism delays the onset and progression of labour 6,7 and mice without oxytocin cannot deliver milk to their young. 5 Hence, oxytocin is required for normal parturition and is essential for milk delivery d… Show more

“…The identification of leader cells within synchronized cell populations that drive episodic pituitary hormone output has remained a major unresolved issue in multiple neuroendocrine systems. For instance, oxytocin neurons of the hypothalamus display synchronized bursts of firing that lead to pulses of oxytocin release from the posterior pituitary gland ( 37 , 38 ). Attempts using techniques such as paired electrophysiological recordings have been unable to determine leader and follower subpopulations ( 39 , 40 ).…”

“…Upon stimulation, magnocellular neurons in the supraoptic (SON) and paraventricular nuclei (PVN) fire high-frequency bursts of action potentials. Each burst generates a large pulse of OT release into the bloodstream to evoke contractions of the uterus and milk ejection [ 46 ]. Likewise, parvocellular neurons release OT toward the central nervous system.…”

“…During pregnancy and parturition, the role of hypothalamic release of Oxt is well discussed ( Veening et al, 2010 ). Hypothalamic Oxt release is also important for lactation ( Perkinson et al, 2021 ). Oxt fibers reach the MEA, multiple regions of the pre-optic area- medial preoptic area (MPO), medial preoptic nucleus (MPN), posterodorsal preoptic nucleus (PD), periventricular hypothalamic nucleus, preoptic part (PVpo), anteroventral preoptic nucleus (AVP) as well as the VMH and PMv.…”
