Vitamin D and MicroRNAs in Bone

Abstract: MicroRNAs (miRNAs) are short noncoding RNAs that orchestrate complex posttranscriptional regulatory networks essential to the regulation of gene expression. Through complementarity with messenger RNA (mRNA) sequences, miRNAs act primarily to silence gene expression through either degradation or inhibited translation of target transcripts. In this way, miRNAs can act to fine-tune the transcriptional regulation of gene expression, but they may also play distinct roles in the proliferation, differentiation, and f… Show more

“…Five miRNAs were found to be differentially expressed in primary human osteoblast after 6 h of treatment with 1,25D3 (Lisse et al, 2013a,b). Interestingly, miR-637 and miR-1228 are two miRNAs located intergenic in DAPK3 and LRP1, respectively.…”

“…miR-637 stimulated the degradation of COL4A mRNA levels that is expressed in the basement membrane and is downregulated during early differentiation of mouse MC3T3-E1 osteoblasts (Hong et al, 2010). It is becoming evident that miRNAs play an important role in osteoblast differentiation and bone formation (Lian et al, 2012) and in the near future more data on their role in 1,25D3 action in osteoblasts will come forward (Lisse et al, 2013a). …”

“…De même, la vitamine D agit sur la régulation mais également le recrutement d'histone acétylases (HAT) et d'histones désacétylases (HDAC), permettant ainsi de jouer sur le niveau d'acétylation des histones (Karlic, et al, 2013). Enfin la vitamine D via le VDR est également capable de réguler l'expression de miRNAs, soit directement soit indirectement en modulant l'activité de facteurs de transcription (Lisse, et al, 2013).…”

“…In addition, recent studies have also implicated microRNAs (miRNAs) in mediating the fine-tuning of vitamin D-mediated responses [2123]. The liganded VDR complex can either suppress or induce miRNAs by either direct transcriptional regulation of autonomous miRNA genesor indirect regulation of miRNAs via host gene promoter sequences [24]. Conversely, miRNAs may act to regulate 1,25(OH) 2 D synthesis, catabolism, or signaling to form dynamic feedback mechanisms (comprehensive review, see [24]).…”
