This work has been prepared to find out changes in the biochemicals with DNA damage, micronucleus, and apoptosis to lipopolysaccharides (LPS) alone or vitamin E (VE) and sodium selenite (SS) in rats’ blood tissue. Rats were divided into eight groups according to the treatment into control vitamin E (VE) treatment group (200 mg/kg bw), sodium selenite (SS) treatment (0.35 mg/kg bw) group, VE + SS treatment group (200 + 0.35 mg/kg bw), LPS treatment group (10 mg/kg bw), LPS + VE (10 + 200 mg/kg bw), LPS + SS treatment (10 + 0.35 mg/kg bw), and LPS + SS+VE treatment (10 + 0.35 + 200 mg/kg bw) group for 6 hr. LPS increased malondialdehyde (MDA) level and decreased antioxidant enzymes’ activities in rat erythrocytes and leukocytes. DNA damage of leukocytes with comet assay and RAPD‐PCR was detected in LPS treatment group. The levels of micronucleus and apoptosis percentage were increased significantly at the end of 6 hr. VE and/or SS protected the LPS‐induced erythrocytes and leukocytes against damage as they have caused amelioration of rats by altering the results. As a result, the co‐administration of VE and/or SS against LPS‐induced damage provides protection. VE and/or SS in patients and animal models with sepsis must be taken in the diet because they are protective against the cellular degradation caused by oxidative damage. Practical applications LPS obtained from E. coli is used more frequently in experimental sepsis studies. When LPS is administered to experimental animals, interstitial pneumonia, adult respiratory fatal syndrome, acute tubular necrosis, and fatal effects such as coagulopathy and hypoglycemia may be seen in these animals. The co‐treatment of VE and SS may be more effective than using them alone against LPS.

Objective: Mercury (Hg) is a highly dangerous and also one of the harmful heavy metals which induces oxidative stress in the animal body. The present study is planned to examine the possible defensive result of caffeic acid (CA) against mercury chloride (HgCl2)-induced hepatotoxicity in male albino Wistar rats, Rattus norvegicus. Methods: Sublethal dose of HgCl2 (1.29 mg/kg body weight) was administrated in rats for 15 days through oral dose and the CA was administrated for another 15 days on mercuric-intoxicated rats. After completing the scheduled exposure time, the rats were sacrificed and the whole liver organ was removed immediately from the animal, and it was used to carry out for biochemical and bioenzymological studies to observe. Results: The level of lipid peroxidation (LPO) content reduced glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) in the liver tissue. CA is an energetic component in the phenolic propolis extract and also in a wide variety of plants, and a strong antioxidant helps to prevent oxidative damage and to reduce oxidative stress. The antioxidants such as GPx, CAT, and SOD and non-antioxidant GSH were significantly decreased, and also, the LPO level was increased in mercury toxicity rats. The treatment of CA (5 mg/kg body weight) in the liver tissue shows considerable declining in the level of oxidant content and along with an increase in the level of antioxidant properties by the way of improvement in liver tissues. Antioxidant and non-antioxidant enzyme (LOP, GSH, GPx, SOD, and CAT) activities were also significantly decreased to near untreated control level when compared to Hg-treated group. The CA acid alone treatment showed the enhanced antioxidant levels and not any alteration in the levels of biochemical parameters when compared with control. Conclusion: These observations of the present experimental study clearly explained the detoxify effects and protective effect of CA against HgCl2 toxicity in liver tissue.

Background: Mercury has a variety of industrial applications and is well-known for its hematotoxic, hepatotoxic, neurotoxic, nephrotoxic, and genotoxic effects. Objectives: This study was carried out to assess the protective effects of gallic acid against mercuric chloride-induced oxidative stress in albino rats. Methods: A total of 35 male Wistar rats were divided into 5 groups (7 rats per group). Groups 1 and 2 were used as the negative and positive controls, respectively and received normal saline (2 mL/kg/day, po) and mercuric chloride (0.4 mg/kg/day, po) for 28 days. Group 3 only received gallic acid (200 mg/kg/day, po) for 28 days, whereas groups 4 and 5 received gallic acid (50 and 200 mg/kg/day, respectively) after 1 hour, followed by mercuric chloride (0.4 mg/kg/day, po) for 28 days. Results:The results demonstrated that treatment with gallic acid significantly diminished the mercuric chloride-induced increase in the serum levels of aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, and lipid peroxidation in liver tissues. In addition, gallic acid treatment increased the level of glutathione peroxidase, superoxide dismutase, and catalase activity and lowered the glutathione level in liver tissues, compared to the mercuric chloride group. The liver of rats, treated with mercuric chloride, showed degenerated cells (with mild cytoplasmic vacuolation and blebbing), binucleated cells, and significant sinusoidal dilation. Conclusions: It can be concluded that gallic acid restores the activities of antioxidant enzymes and tissue markers in mercuric chloride-treated rats, probably by scavenging free radicals and improving the antioxidant defense mechanisms.
