VLA-4-Mediated Interactions Between Normal Human Hematopoietic Progenitors and Stromal Cells

Abstract: The alpha 4 beta 1 integrin very late activation antigen-4 (VLA-4) has been implicated to play a role in the adhesive interactions between hematopoietic progenitor cells (HPC) and bone marrow stromal cells which express the vascular cell adhesion molecule-1 (VCAM-1) or produce fibronectin (FN). Here, we summarize some of the recent advances made in the elucidation of the role of these particular adhesive interactions for the regulation of normal hematopoiesis. HPC bind to stroma mainly through VLA-4/VCAM-1 int… Show more

“…Until now, most investigators have attempted to elucidate the impact of a single angiogenic factor on the pathogenesis or prognosis of haematologic malignancies (Aguayo et al, 2002;Litwin et al, 2002;Moehler et al, 2003;Liu et al, 2005;Nowicki et al, 2006). However, HSCs were observed to be in close contact with the marrow stromal cells in vivo (Oostendorp and Dormer, 1997;Tordjman et al, 1999). Furthermore, both fibroblasts and microvascular ECs obtained in normal and leukaemic marrows were reported to produce several angiogenesis molecules (Moehler et al, 2003;De Raeve et al, 2004;Ogawa et al, 2006).…”

“…It is expressed on human CD34ϩ cells, and murine HSPCs (31)(32)(33). Blocking the interaction between VLA-4 and its ligands using anti-VLA-4-specific antibodies, or small molecule inhibitors induces mobilization of HSPCs in humans (34,35), primates (36,37), and mice (38).…”

“…This regulation appears to be mediated by mutual recognition between adhesion molecules expressed by stromal and vascular endothelial cells (ECs) of hematopoietic tissues and their ligands on HSPCs [26,29,35,37]. Moreover, it is well known that cell adhesion molecules and ligands regulate the migration of HSPCs from the blood stream into the hematopoietic compartment [27].…”
