Voltage-dependent ionic currents in solitary horizontal cells isolated from cat retina

Abstract: 1. Horizontal cells of the cat retina were isolated by enzymatic dissociation. Two types of horizontal cells were identified: the axonless (A-type) horizontal cell having four to six thick, long (approximately 100 microns) dendrites, and the short-axon (B-type) horizontal cell having many (> 5) fine, short (approximately 30 microns) dendrites. 2. Membrane properties of isolated horizontal cells were analyzed under current-clamp and voltage-clamp conditions. In the A-type cell, the average resting potential was… Show more

“…Dissociated bipolar cells were prepared according to the procedures described previously (Ueda et al, 1992), with some modifications. Eyes from pups of postnatal days (PD) 12-20 were enucleated, and the sclera and choroid were peeled off.…”

“…Voltage-gated sodium channels have been identified on isolated horizontal cells (Shingai and Christensen 1983;Ueda et al 1992) and on human rod photoreceptors (Kawai et al 2001), but there is no evidence that sodium currents shape light responses of these neurons. If salamander photoreceptors or horizontal cells express voltage-gated sodium channels, then TTX could alter transmission from photoreceptors to secondorder neurons.…”

“…There are reports of voltage-gated sodium channels in certain classes of photoreceptors, horizontal cells, and cone bipolar cells in other species (Kawai et al 2001;Pan and Hu 2000;Shingai and Christensen 1983;Ueda et al 1992;Zenisek et al 2001). In the salamander, these channels have been observed on ganglion and amacrine cells but never on other retinal neurons (Barnes and Werblin 1986;Lukasiewicz and Werblin 1988).…”

“…Single bipolar cells were isolated from retinas of pink-eyed RCS rats between postnatal day 25 and 55 and of normal pigmented rats according to the procedure used by Ueda et al [1992] with minor modifications. 500 µl of the cell suspension were transferred into a plastic culture dish previously filled with 2 ml of the standard extracellular solution which contained (in mM): 10 KCl, 130 NaCl, 1 MgCl 2 , 2 CaCl 2 , 5 N-2-hydroxyethylpiperazine-N′-2-ethane-sulfonic acid (HEPES), and 10 glucose.…”
