VWF, Platelets and the Antiphospholipid Syndrome

Abstract: The antiphospholipid syndrome (APS) is characterized by thrombosis and/or pregnancy morbidity with the persistent presence of antiphospholipid antibodies (aPLs). Laboratory criteria for the classification of APS include the detection of lupus anticoagulant (LAC), anti-cardiolipin (aCL) antibodies and anti-β2glycoprotein I (aβ2GPI) antibodies. Clinical criteria for the classification of thrombotic APS include venous and arterial thrombosis, along with microvascular thrombosis. Several aPLs, including LAC, aβ2GP… Show more

“…The pathogenesis of thrombosis in APS is complex, multifactorial and is still not fully understood [ 25 , 26 , 27 ]. What is clear is that thrombosis is the hallmark of the disease, and that multiple mechanisms involving endothelial cells, platelets, monocytes, neutrophils, and the coagulation and complement pathway are involved.…”

“…Wan et al indicated that LA positivity was an independent associated factor with brain MRI abnormalities (especially lacunae and WMHs) in people without neuropsychiatric symptoms ( 39 ), However, in our study, we tested lupus patients with IgG/IgM aβ 2 -GPI (targeting β 2 -GPI Domain I) and IgG/IgM aCL and did not further investigate IgA aβ 2 -GPI (targeting β 2 -GPI Domain III IV V) or identify β 2 -GPI-dependent aCL IgG. Their diagnostic and clinical significance have been well demonstrated in other studies, which indicates they are correlated with arterial thrombosis ( 26 , 40 , 41 ). This issue will be further explored in our future research.…”

“…In this study we investigated a broad range of specific haemostatic parameters in SARS-CoV-2 PCR positive patients presenting at the emergency department during the first period of the SARS-CoV-2 pandemic. We found that SARS-CoV-2 PCR positive patients had a prothrombotic phenotype as seen by higher fibrinogen and VWF levels [ 25 ], and acquired activated protein C resistance [ 26 ]. One out of seven hospitalized PCR positive patients developed thrombosis, which corresponds well to prevalence of thrombosis in COVID-19 patients reported in other studies [ 27 ].…”
