Ways to Address Perinatal Mast Cell Activation and Focal Brain Inflammation, including Response to SARS-CoV-2, in Autism Spectrum Disorder

Abstract: The prevalence of autism spectrum disorder (ASD) continues to increase, but no distinct pathogenesis or effective treatment are known yet. The presence of many comorbidities further complicates matters, making a personalized approach necessary. An increasing number of reports indicate that inflammation of the brain leads to neurodegenerative changes, especially during perinatal life, “short-circuiting the electrical system” in the amygdala that is essential for our ability to feel emotions, but also regulates … Show more

“…We then manually searched PubMed for articles within our 598 that were related to Covid-19, and found 15 relevant papers (Table 4). Six of them are reviews [37][38][39][40][41][42], two studies provide perhaps the clearest evidence on the subject [43,44]; one is a comparative study that perhaps provides the clearest evidence on the subject [44]. It is a retrospective report on 311 patients with severe Covid-19, treated in February of 2020 at the Wuhan Union Hospital, China.…”

“…Other phenotypes such as those with immune deficiencies as well as those with autoimmune encephalopathies are also described. In another article, the importance of mast cell activation in activating microglia resulting in focal brain inflammation and disrupted synaptic pruning is discussed [11]. Similar to EDS/HSD, aberrant mast cell activation and related symptomatology are common in cohorts with ASD and may designate endophenotypes of diseases.…”

“…Two papers discuss potential treatments for immune dysfunction in ASD. Using a systematic review and meta-analysis approach, evidence for the effectiveness of intravenous immunoglobulin (IVIG) in ASD is presented [10], while another study discusses approaches to treating mast cell disorders with an emphasis on luteolin [11].…”
