Wen‐Dan Decoction Improves Negative Emotions in Sleep‐Deprived Rats by Regulating Orexin‐A and Leptin Expression

Abstract: Wen-Dan Decoction (WDD), a formula of traditional Chinese medicine, has been clinically used for treating insomnia for approximately 800 years. However, the therapeutic mechanisms of WDD remain unclear. Orexin-A plays a key role in the sleep-wake cycle, while leptin function is opposite to orexin-A. Thus, orexin-A and leptin may be important factors in sleep disorders. In this study, 48 rats were divided into control, model, WDD-treated, and diazepam-treated groups. The model of insomnia was produced by sleep … Show more

“…Furthermore, WDD has been widely applied in psychological medicine and stroke (Wu et al, 1999;Gao, 2000;Chun et al, 2003). Recently, WDD treatment normalized the behavior and up-regulated orexin-A, leptin, orexin receptor 1 and leptin receptor in the brain (Wu et al, 2014). All these findings indicate the important roles of psychological factors in digestive disorders.…”

“…Increased levels of hcrt-1 in humans have been found during social interactions and in connection with social-induced positive emotions [ 11 ]. Indeed, hcrt neurons receive abundant input from the limbic system [ 33 ] and several papers have provided experimental evidence for an involvement of the hcrt system in emotion and emotional memory [ 28 , 34 , 35 ]. It is often speculated that that activation of hcrt neurons by the limbic system maintains wakefulness during emotional arousal.…”

“…At present, adjuvant drugs for SD play a role in the central nervous system, such as amphetamine, caffeine, and modafinil [ 13 ]. These medications are often associated with a number of side effects, including excessive sleepiness, thought disorders, nocturnal wandering, agitation, and balance problems, thereby limiting clinical use [ 14 ]. Ginseng roots ( Panax ginseng C. A Meyer), one of the best known traditional Chinese medicine (TCM) herbs, have been reported to display adaptogenic effects in the endocrine, immune, cardiovascular, and central nervous systems [ 15 ].…”
