Wheat germ lipase: isolation, purification and applications

Abstract: In recent years, wheat germ lipase (WGL) is attracting considerable interest. To date, several WGL applications have already been described: (i) fats and oils modification; (ii) esterification reactions in organic media, accepting a wide range of acids and alcohols as substrates; (iii) the asymmetric resolution of various chiral racemic intermediates; (iv) more recently, the promiscuous activity of WGL has been shown in carbon-carbon bond formation. To date, no crystallographic structure of this enzyme has bee… Show more

“…In the experiments on lipase-treated granum and stroma TMs, a substrate non-specific [44], general tri-, di-, and monoglyceride hydrolase-lipase from wheat germ (L3001, Sigma-Aldrich, Burlington, MA, USA) was used. A stock solution of 0.5 U µL −1 activity was prepared in MilliQ water, and volumes containing the desired activity of the lipase were added into the sample and thoroughly mixed prior to the start of the measurement.…”

“…Two types of lipases were used and characterized in detail regarding their effects on the lipid polymorphism of TMs: Phospholipase A1 (PLA1), which selectively cleaves off the fatty acid on the sn-1 position from the glycerol backbone [ 51 ], and Wheat-Germ Lipase (WGL), which is a substrate non-specific/promiscuous lipid hydrolase [ 52 ].…”

“…Goose, chicken, bovine, wild hog and deer livers were converted to the acetone powder (GLAP) by the method of Connors et al [ 77 ]. Homemade lipase from wheat germ was prepared according to the literature protocol [ 78 ]. Merck silica gel plates 60 F254 were used for TLC (Thin Layer Chromatography) analysis.…”
