Whole-genome integrative analysis reveals expression signatures predicting transformation in follicular lymphoma

Abstract: Key Points Whole-genome integrative analyses in FL reveal that genes strongly influenced by copy number are highly enriched for NF-kB pathway regulators. Subsignatures of the NF-kB targets predict transformation in FL.

“…22 Similarly, the observation that a small gene expression panel based on components of the nuclear factor kB (NF-kB) pathway components is predictive of HT may directly reflect the recent sequencing data showing that HT is frequently accompanied by mutations in genes encoding components of the NF-kB pathway, as mutations affecting the NF-kB pathway appear to be among the most consistent of the stepwise changes that ultimately lead to clinically recognizable HT. 3,23 Summary: biology of transformation…”

“…A Phase I study [30] revealed a response rate of 36% in follicular lymphoma, as compared with 69-75% in the CLL, Waldenström's macroglobulinemia, and MCL histology subgroups. The ultimate course of follicular lymphoma, namely its transformation into aggressive NHL, is associated with the upregulation of NF-kB regulating genes, including BTK [38,39]. However, as transformed follicular lymphoma patients are often excluded from follicular lymphoma trials, no data on ibrutinib therapy in this particular setting is available to date.…”

“…Immune response 1 profile was found to be associated with a favorable survival prediction compared to the second immune response profile [48]. Recent analyses of copy number alterations (CNA) and gene mutation have provided greatly increased information on the pathogenesis and progression of FL [49][50][51]. This includes frequent abnormalities of genes involved in chromatin modification.…”
