Micrococcus luteus is a predominant member of skin microbiome. We here report on the genomic analysis of Micrococcus luteus strain O'Kane that was isolated from an elevator. The partial genome assembly of Micrococcus luteus strain O'Kane is 2.5 Mb with 2256 protein-coding genes and 62 RNA genes. Genomic analysis revealed metabolic versatility with genes involved in the metabolism and transport of glucose, galactose, fructose, mannose, alanine, aspartate, asparagine, glutamate, glutamine, glycine, serine, cysteine, methionine, arginine, proline, histidine, phenylalanine, and fatty acids. Genomic comparison to other M. luteus representatives identified the potential to degrade polyhydroxybutyrates, as well as several antibiotic resistance genes absent from other genomes.

A total of eight varieties of the mango from an orchard were studied using molecular markers to understand the host-pathogen interaction. From the infected leaves of the plant, a total of the 8 bacterial pathogens (Exiguobacterium arabatum, Pseudomonas mendocina, Pantoea dispersa, Bacillus sp. Pantoea ananatis, Micrococcous luteus, Microbacterium_sp., Enterobacter cloacae) were isolated and identified. All the host varieties of mango were distinguished for the genetic diversity using the Inter Simple Sequence Repeat (ISSR) DNA markers. This set of ISSR marker primers were also used for the mango pathogens. PCR amplification of the ISSR primers showed polymorphic and monomorphic band patterns in the host plants and in their pathogens. The monomorphic band generated by PCR amplification in the host and in the pathogen, by the common primer, is selected and used for PCR hybridization technique. PCR products obtained from the host, pathogen and hybridization were cloned, sequenced and compared. A multiple sequence alignment of these sequences revealed that the product of hybridization PCR was mixture of host and pathogen sequences. On this basis, we hypothesize a possibility for the recombination of host-microbes DNA as one of the mechanisms of pathogenicity for the plant pathogens using hybrid PCR technique. The possible mechanism of recombination for plant host and its pathogen is discussed.HighlightsInter Simple Sequence Repeat markers used to (i) Fingerprint the pathogens and their host (mango) and (ii) for study of the possibilities for the recombination as mechanism of pathogenicity.

Detection of seed-borne pathogenic bacteria of northern black wattle (Acacia auriculiformis A. Cunn. ex Benth.). Intensive research of seed-borne pathogen of A. mangium and A. crassicarpa which have been established in industrial timber estate (HTI), while plantings development of northern black wattle in Indonesia 1990 have not recent been established in the 1990s. Very limited information available on northern black wattle diseases especially seed-borne diseases. The objectives of this study were to identify seed-borne pathogenic bacteria of northern black wattle and the effects on seed germination. Methods for the isolation of bacteria were by seed soaking, seed griding, blotter test, growing-on test on paper and soil. Identification of bacteria by PCR used 63F/1387R primer. The results showed that seed-borne bacteria of northern black wattle were Paenochrobactrum sp., Ralstonia sp., Burkholderia cepacia complex, Pseudomonas stutzeri, Acinetobacter sp., Alcaligenes faecalis, Salmonella bongori, Escherichia hermannii while pathogenic bacteria cause seedling leafspot were Micrococcus luteus and Burkholderia cepacia complex. Burkholderia cepacia complex, A. faecalis, Acinetobater sp., P. stutzeri, S. bongori and Ralstonia sp. reduced seed germination and increased rotten seed, suggested that they were the pathogenic bacteria of northern black wattle seed. Ralstonia sp. significantly increased the percentage of rotten seed and decreased shoot length and root length. P. stutzeri and S. bongori significantly inhibited the root growth. Paenochrobactrum sp. and E. hermannii were assumed as pathogen with weak virulence due to seed germination, the percentage of rotten seed and vigour index were relatively similar to untreated seed.
