Would You Use It With a Seal of Approval? Important Attributes of 2,4-Dinitrophenol (2,4-DNP) as a Hypothetical Pharmaceutical Product

Abstract: Background2,4-Dinitrophenol (2,4-DNP) is an effective but highly dangerous fat burner, not licensed for human consumption. Death cases reported for 2,4-DNP overdose, particularly among young adults, have raised concerns about the ineffective regulatory control, lack of education and risks associated with impurity, and the unknown concentration of 2,4-DNP purchased on the Internet.MethodsUsing a sequential mixed method design and based on a hypothetical scenario as if 2,4-DNP was a licensed pharmaceutical drug,… Show more

“…In addition, once pharmacologic agents act to decrease PMF, they cannot be reversed. Therefore, using drugs to decrease the PMF is dangerous due to narrow therapeutic windows and lack of tissue selectivity 19–21 . This difficulty in developing PMF therapy highlights the need for spatiotemporal control in experimental models to understand how decreasing the PMF provides stress resistance.…”

“…Therefore, using drugs to decrease the PMF is dangerous due to narrow therapeutic windows and lack of tissue selectivity. [19][20][21] This difficulty in developing PMF therapy highlights the need for spatiotemporal control in experimental models to understand how decreasing the PMF provides stress resistance. How and where the metabolic state of an organism is sensed, and how that translates to physiologic changes remains unclear.…”

“…The consistent low ranking of "excellence is performance" is a perplexing outcome which warrants further exploration. From the research point of view, values in the middle ranks also present an intriguing question, namely if the reason for a more neutral ranking is caused by indifference, or more likely, large disagreement at the individual level as observed previously with the BWS method (Bleasdale et al, 2018).…”

“…At the same time, this process breaks down carbohydrates and fats, allowing energy from cellular respiration to be released as heat. The excess heat production led to uncontrolled toxic hyperthermia associated with significant morbidity and mortality 6) . The history of DNP becomes an important lesson that clinical application of a mitochondrial uncoupler needs finetuned dosing regimens that spare adverse effects.…”
