The possible importance of the cysteine-rich γ-prolamin in kafirin and zein functionality has been neglected. Gamma-kafirin"s role in organized microstructures was investigated in microparticles. Residual kafirin (total kafirin minus -kafirin) "microparticles" were nondiscrete (amorphous mass of material), as viewed by electron microscopy and atomic force microscopy. Adding 15% -kafirin to residual kafirin resulted in formation of a mixture of non-discrete material and nanosize discrete spherical structures. Adding 30% -kafirin to the residual kafirin resulted in discrete spherical nanosize particles. FTIR indicated that γ-kafirin had a mixture of random coil and β-sheet conformation, in contrast to total kafirin which is mainly α-helical. Gamma-kafirin also had a very high glass transition temperature (T g ) (270 o C). The conformation and high T g of γ-kafirin probably confer structural stability to kafirin microstructures. Due to its ability to form disulfide cross-links, gamma-kafirin appears to be essential to form and stabilize organized microstructures.

Influence of Formulation Factors on the Preparation of Zein Nanoparticles

Abstract. The main objective of the present study was to investigate the influence of various formulation parameters on the preparation of zein nanoparticles. 6,7-dihydroxycoumarin (DHC) was used as a model hydrophobic compound. The influence of pH of the aqueous phase, buffer type, ionic strength, surfactant, and zein concentration on particle size, polydispersity index, and zeta potential of DHC-loaded zein nanoparticles were studied. Smaller nanoparticles were formed when the pH was close to the isoelectric point of zein. DHC-loaded zein nanoparticles prepared using citrate buffer (pH 7.4) was better than phosphate buffer in preventing particle aggregation during lyophilization. The ionic strength did not have a significant influence on the particle size of DHC-loaded zein nanoparticles. A combination of Pluronic F68 and lecithin in 2:1 ratio stabilized the zein nanoparticles. An increase in zein concentration led to increase in particle size of DHC-loaded zein nanoparticles. The use of optimal conditions produced DHCloaded nanoparticles of 256±30 nm and an encapsulation efficiency of 78±7%. Overall, the study demonstrated the optimal conditions to prepare zein nanoparticles for drug encapsulation.

Zein-based nano/microparticles have been demonstrated to be promising carrier systems for both the food industry and biomedical applications. However, the fabrication of size-controlled zein particles has been a challenging issue. In this study, a modified anti-solvent precipitation method was developed, and the effects of various factors, such as mixing method, solvent/anti-solvent ratio, temperature, zein concentrations and the presence of sodium caseinate (SC) on properties of zein particles were investigated. Evidence is presented that, among the previously mentioned factors, the mixing method, especially mixing rate, could be used as an effective parameter to control the size of zein particles without changing other parameters. Moreover, through fine-tuning the mixing rate together with zein concentration, particles with sizes ranging from nanometers to micrometers and low polydispersity index values could be easily obtained. Based on the size-controlled fabrication method, SC-coated zein nanoparticles could also be obtained in a size-controlled manner by incubation of the coating material with the already-formed zein particles. The resultant nanoparticles showed better performance in both drug loading and controlled release, compared with zein/SC hybrid nanoparticles fabricated by adding aqueous ethanol solution to SC solution. The possible mechanisms of the nanoprecipitation process and self-assembly formation of these nanoparticles are discussed.
