Zeta potential vs apoptotic marker: which is more suitable for ICSI sperm selection?

Abstract: Both MACS and Zeta procedures improve the quality of the selected spermatozoa for ICSI. However, MACS procedure is more efficient in individuals with severe male factor infertility to select sperm with normal acrosome and protamine content but concern regarding transfer of MACS beads into the oocyte remains to be resolved.

“…The identification and elimination of “apoptotic” spermatozoa, those showing phosphatidylserine (PS) transposition, is a relatively common procedure in human fertility clinics. It is believed that the elimination of these apoptotic spermatozoa using magnetic beads increases fertility (Zahedi et al., ). The membrane of the mammalian spermatozoa can experience damage that is not detected using classical dye exclusion assays.…”

“…Externalization of PS from inner to outer layer of plasma membrane can be assessed by Annexin V and is considered as one of the early signs of apoptosis before capacitation. [43] However, this process is a nor- Steps of acrosome reaction mal phenomenon observed during capacitation. This dual functioning role of PS in human spermatozoa makes interpretation and test design very difficult.…”

“…It has been emphasized that the MACS assay has the potential to isolate spermatozoa with normal acrosomes and protamine content and to have a higher significance in severe teratozoospermic patients. [46] Zeta sperm selection A mature sperm possess a surface electric charge ranging from -16 to -20 mV, termed as the zeta potential which decreases with capacitation. [47] Normally differentiated sperms are charged electronegatively ( Figure 7).…”

“…Slides were observed under a microscope (Zeiss, Oberkochen, Germany) equipped with a 100-magnification lens. [54][55][56][57][58][59][60] Spermatozoa with fragmented DNA had browncolored nuclei, whereas the other cells were blue-gray (counter coloration with Harris's haematoxylin) [7,[61][62][63]. On each slide, approximately 500 cells were counted, and the DNA fragmentation index (DFI) was calculated ( Figure 2).…”
