Zika virus as a causative agent for primary microencephaly: the evidence so far

Abstract: Zika virus (ZIKV) infection has been associated with congenital microcephaly and peripheral neuropathy. The ongoing epidemic has triggered swift responses in the scientific community, and a number of recent reports have now confirmed a causal relationship between ZIKV infection and birth defect. In particular, ZIKV has been shown to be capable of compromising and crossing the placental barrier and infect the developing fetal brain, resulting in the demise and functional impairment of neuroprogenitor cells crit… Show more

“…As a result, our work emphasizes the need to focus research efforts on unraveling the true source of these microcephaly cases, keeping in mind that it may involve a complex combination of causes. As the ZIKV infection is now becoming more and more strongly correlated with the emergence of these microcephaly phenotypes42525354, we recommend that the scientific community continue to pursue research along this path.…”

“…We examined three different HEK293-derivatives, HEK293-FIT, HEK293-T3Y and 293T cells, and found they supported varying degrees of ZIKV replication (see below). A notable feature of ZIKV infection in humans is its neurotropism [46][47][48]. We thus examined two human cell lines of neural origin, i.e., the neuroblastoma line SK-N-SH and SVGA, a fetal glial cell line, and found both to be permissive for ZIKV infection (S3 Fig).…”

“…In pregnant women, the disease can cause fetal brain damage and microcephaly, whereas in adults it causes Guillain-Barré syndrome. Zika virus was also found in semen, and sexual transmission has been reported 34 . The attack rate is estimated at 14.6/1,000 inhabitants during epidemics; however, the exact risk for travelers remains unknown 35 .…”
