Experimental Studies on the Diagnosis of Marginal Zinc DeficiencySummary. A hundred and twelve male Sprague-Dawley rats with an average live weight of 37 g were depleted for 14 days with a Zn-deficient diet of 1.3 mg Zn/kg. Thereafter, they were divided into 16 groups of 7 animals each. For another 14 days, two groups each were given diets containing 1.3, 4, 6, 8, 10, 12, 20 and 100ppm Zn. Then one of each of these groups were sacrificed, while the other received a Zn injection of 0.8 mg Zn/animal. Three days later, these 8 groups were also sacrificed in order to test the effect of Zn injection on several parameters of Zn supply.1. The Zn requirement for an optimum growth of the young rats was reached at 12 ppm. 2. The serum and bone contents increased in response to Zn injection up to the dietary supply of 20 ppm Zn; the Zn content of the testes was not affected. 3. The activity of the alkaline phosphatase in serum showed a significant increase after Zn injection up to those animals given 20 ppm dietary zinc and remained unchanged at 100ppm. 4. In bone and testes the activity of the alkaline phosphatase increased after Zn injection up to the groups with 8 and 10 ppm dietary Zn, respectively.The activity of the alkaline phosphatase in serum and its response to Zn injection thus represents a simple and sensitive system to assess the Zn supply status, especially the marginal supply. If an increase in the activity of the serum alkaline phosphatase can be obtained by Zn injection, a more or less pronounced Zn undernutrition, depending upon the observed activity increase, is indicated because no increase in the activity could be effected by additional Zn doses when the Zn status was optimal. 1. Der Zn-Bedarf ffir ein optimales Wachstum der jungen Ratten war bei 12 ppm erreicht. 2. Serum-und Knochen-Zn-Gehalte erh6hten sich nach einer Zn-Injektion bis zu einem Di~it-Zn-Gehalt von 20ppm und blieben im Hoden bei allen Gruppen unbeeinflugt. 3. Die Aktivitfit der alkalischen Phosphatase im Serum zeigte auf die ZnInjektion eine signifikante Aktivit~itserh6hung bis zu den Tieren mit einem Di~it-Zn-Gehalt von 20ppm. Sie blieb bei 100ppm unver~indert. 4. Im Knochen und Hoden erh6hte sich die Aktivit~it der alkalischen Phosphatase nach einer Zn-Injektion bis zu den Gruppen mit 8 bzw. 10 ppm DifitZn-Gehalt.Die Aktivit~it der alkalischen Phosphatase im Serum und deren Respons infolge einer Zn-Injektion stellt damit ein einfaches und sensitives System zur Bestimmung des Zn-Versorgungsstatus, speziell im marginalen Bereich, dar. Lfigt sich aufgrund einer Zn-Injektion eine Aktivit~itserh6hung der alkalischen Phosphatase im Serum erzielen, so mug je nach der H6he der festgestellten Aktivitfitserh6hung eine mehr oder weniger ausgeprfigte Zn-Unterversorgung vorgelegen haben, da bei einer optimalen Versorgung sich durch zusfitzliche Zn-Gaben keine Aktivit~ttserh6hung erzielen lieg.Schliisselwiirter: Zn-Mangel -Diagnose von Zn-Mangel -alkalische Phosphatase -saure Phosphatase -Zn-Injektion -Serum-Zn -Femur-ZnHoden-Zn
