Δ 5 -3β-Hydroxysteroid dehydrogenase from Digitalis lanata Ehrh. - a multifunctional enzyme in steroid metabolism?

Abstract: Delta(5)-3beta-Etaydroxysteroid dehydrogenase (Delta(5)-3beta-HSD; EC 1.1.1.145), an enzyme converting pregn-5-ene-3beta-ol-20-one (pregnenolone) to pregn-5-ene-3,20-dione (isoprogesterone), was isolated from the soluble fraction of suspension-cultured cells of Digitalis lanata L. strain VIII. Starting with acetone dry powder the enzyme was purified in three steps using column chromatography on Fractogel-TSK DEAE, hydroxyapatite and Sephacryl G-200. Fractions with highest Delta(5)-3beta-HSD activity were separ… Show more

“…This protein contains 3␤-HSD and NmrA components. 3␤-HSD has been implicated in the biosynthesis of the steroid cardenolide in foxglove (Digitalis lanata) (69) and has been detected in potato and tobacco, suggesting a role in general steroid metabolism (70). In a proteomics survey of Panax ginseng, the abundance of 3␤-HSD increased in response to light and was linked to the biosynthesis of steroidal saponins (71).…”

“…The highest similarities were seen with sequences from other plants (Scherbak et al, 2009): (2)-isopiperitenol dehydrogenase from Mentha species (similarity of 55%; Ringer et al, 2005), D 5 -3b-hydroxysteroid dehydrogenase (HSD; EC 1.1.1.145) from Digitalis lanata (49%; Finsterbusch et al, 1999), and secoisolariciresinol dehydrogenase from Podophyllum peltatum (46%; Xia et al, 2001). The threedimensional structure of the Podophyllum protein has been solved (Youn et al, 2005), while the Mentha protein is the polypeptide with the highest similarity to the pea SADs for which an enzymatic function is known.…”
