BackgroundLight curtain arrays (LC), a recently introduced phenotyping method, yield a binary data matrix from which a shoot silhouette is reconstructed. We addressed the accuracy and applicability of LC in assessing leaf area and maximum height (base to the highest leaf tip) in a phenotyping platform. LC were integrated to an automated routine for positioning, allowing in situ measurements. Two dicotyledonous (rapeseed, tomato) and two monocotyledonous (maize, barley) species with contrasting shoot architecture were investigated. To evaluate if averaging multiple view angles helps in resolving self-overlaps, we acquired a data set by rotating plants every 10° for 170°. To test how rapid these measurements can be without loss of information, we evaluated nine scanning speeds. Leaf area of overlapping plants was also estimated to assess the possibility to scale this method for plant stands.ResultsThe relation between measured and calculated maximum height was linear and nearly the same for all species. Linear relations were also found between plant leaf area and calculated pixel area. However, the regression slope was different between monocotyledonous and dicotyledonous species. Increasing the scanning speed stepwise from 0.9 to 23.4 m s−1 did not affect the estimation of maximum height. Instead, the calculated pixel area was inversely proportional to scanning speed. The estimation of plant leaf area by means of calculated pixel area became more accurate by averaging consecutive silhouettes and/or increasing the angle between them. Simulations showed that decreasing plant distance gradually from 20 to 0 cm, led to underestimation of plant leaf area owing to overlaps. This underestimation was more important for large plants of dicotyledonous species and for small plants of monocotyledonous ones.ConclusionsLC offer an accurate estimation of plant leaf area and maximum height, while the number of consecutive silhouettes that needs to be averaged is species-dependent. A constant scanning speed is important for leaf area estimations by using LC. Simulations of the effect of varying plant spacing gave promising results for method application in sets of partly overlapping plants, which applies also to field conditions during and after canopy closure for crops sown in rows.
Solar radiation is a key environmental signal in regulation of plant secondary metabolism. Since metabolic responses to light and ultraviolet (UV) radiation exposure are known to depend on the ratio of spectral ranges (e.g., UV-B/PAR), we examined effects of different UV-B radiation (280-315 nm) and photosynthetically active radiation (PAR, 400-700 nm) levels and ratios on yield and pattern of monoterpenoid essential oil of peppermint. Experiments were performed in exposure chambers, technically equipped for realistic simulation of natural climate and radiation. The experimental design comprised four irradiation regimes created by the combination of two PAR levels including or excluding UV-B radiation. During flowering, the highest essential oil yield was achieved at high PAR (1150 micromol m(-2) s(-1)) and approximate ambient UV-B radiation (0.6 W m(-2)). Regarding the monoterpene pattern, low PAR (550 micromol m(-2) s(-1)) and the absence of UV-B radiation led to reduced menthol and increased menthone contents and thereby to a substantial decrease in oil quality. Essential oil yield could not be correlated with density or diameter of peltate glandular trichomes, the epidermal structures specialized on biosynthesis, and the accumulation of monoterpenes. The present results lead to the conclusion that production of high quality oils (fulfilling the requirements of the Pharmacopoeia Europaea) requires high levels of natural sunlight. In protected cultivation, the use of UV-B transmitting covering materials is therefore highly recommended.
In order to produce protein-rich duckweed for human and animal consumption, a stable cultivation process, including an optimal nutrient supply for each species, must be implemented. Modified nutrient media, based on the N-medium for duckweed cultivation, were tested on the relative growth rate (RGR) and crude protein content (CPC) of Lemna minor and Wolffiella hyalina, as well as the decrease of nitrate-N and ammonium-N in the media. Five different nitrate-N to ammonium-N molar ratios were diluted to 10% and 50% of the original N-medium concentration. The media mainly consisted of agricultural fertilizers. A ratio of 75% nitrate-N and 25% ammonium-N, with a dilution of 50%, yielded the best results for both species. Based on the dry weight (DW), L. minor achieved a RGR of 0.23 ± 0.009 d−1 and a CPC of 37.8 ± 0.42%, while W. hyalina’s maximum RGR was 0.22 ± 0.017 d−1, with a CPC of 43.9 ± 0.34%. The relative protein yield per week and m2 was highest at this ratio and dilution, as well as the ammonium-N decrease in the corresponding medium. These results could be implemented in duckweed research and applications if a high protein content or protein yield is the aim.
Root temperature has long been considered an essential environmental factor influencing the plant’s physiology. However, little is known about the effect of root temperature on the quality of the food produced by the plant, especially that of horticultural crops. To fill this gap, two independent root cooling experiments (15 °C vs. 20 °C and 10 °C vs. 20 °C) were conducted in autumn 2017 and spring 2018 in hydroponics with Chinese broccoli (Brassica oleracea var. alboglabra Bailey) under greenhouse conditions. The aim was to investigate the effect of root temperature on plant growth (biomass, height, yield) and food quality (soluble sugars, total chlorophyll, starch, minerals, glucosinolates). A negative impact on shoot growth parameters (yield, shoot biomass) was detected by lowering the root temperature to 10 °C. Chinese broccoli showed no response to 15 °C root temperature, except for an increase in root biomass. Low root temperature was in general associated with a higher concentration of soluble sugars and total chlorophyll, but lower mineral levels in stems and leaves. Ten individual glucosinolates were identified in the stems and leaves, including six aliphatic and four indolic glucosinolates. Increased levels of neoglucobrassicin in leaves tracked root cooling more closely in both experiments. Reduction of root temperature by cooling could be a potential method to improve certain quality characters of Chinese broccoli, including sugar and glucosinolate levels, although at the expense of shoot biomass.
