BackgroundEfficiency of feed utilization is important for animal production because it can reduce greenhouse gas emissions and improve industry profitability. However, the genetic basis of feed utilization in livestock remains poorly understood. Recent developments in molecular genetics, such as platforms for genome-wide genotyping and sequencing, provide an opportunity to identify genes and pathways that influence production traits. It is known that transcriptional networks influence feed efficiency-related traits such as growth and energy balance. This study sought to identify differentially expressed genes in animals genetically divergent for Residual Feed Intake (RFI), using RNA sequencing methodology (RNA-seq) to obtain information from genome-wide expression profiles in the liver tissues of Nelore cattle.ResultsDifferential gene expression analysis between high Residual Feed Intake (HRFI, inefficient) and low Residual Feed Intake (LRFI, efficient) groups was performed to provide insights into the molecular mechanisms that underlie feed efficiency-related traits in beef cattle. A total of 112 annotated genes were identified as being differentially expressed between animals with divergent RFI phenotypes. These genes are involved in ion transport and metal ion binding; act as membrane or transmembrane proteins; and belong to gene clusters that are likely related to the transport and catalysis of molecules through the cell membrane and essential mechanisms of nutrient absorption. Genes with functions in cellular signaling, growth and proliferation, cell death and survival were also differentially expressed. Among the over-represented pathways were drug or xenobiotic metabolism, complement and coagulation cascades, NRF2-mediated oxidative stress, melatonin degradation and glutathione metabolism.ConclusionsOur data provide new insights and perspectives on the genetic basis of feed efficiency in cattle. Some previously identified mechanisms were supported and new pathways controlling feed efficiency in Nelore cattle were discovered. We potentially identified genes and pathways that play key roles in hepatic metabolic adaptations to oxidative stress such as those involved in antioxidant mechanisms. The Cornell Net Carbohydrate and Protein System (CNCPS) model has been increasingly used in tropical regions for dairy and beef production. However, the lack of appropriate characterization of the feeds has restricted its application. The objective of this study was to develop and evaluate a feed library containing feeds commonly used in tropical regions with characteristics needed as inputs for the CNCPS. Feed composition data collected from laboratory databases and from experiments published in scientific journals were used to develop this tropical feed library. The total digestible nutrients (TDN) predicted at 1x intake of maintenance requirement with the CNCPS model agreed with those predicted by the Weiss et al. (1992) equation (r² of 92.7%, MSE of 13, and bias of 0.8%) over all feeds. However, the regression r² of the tabular TDN values and the TDN predicted by the CNCPS model or with the Weiss equation were much lower (58.1 and 67.5%, respectively). A thorough comparison between observed and predicted TDN was not possible because of insufficient data to characterize the feeds as required by our models. When we used the mean chemical composition values from the literature data, the TDN predicted by our models did not agree with the measured values. We conclude using the TDN values calculated using the Weiss equation and the CNCPS model that are based on the actual chemical composition of the feeds result in energy values that more accurately represent the feeds being used in specific production situations than do the tabular values. Few papers published in Latin America journals that were used in this study reported information need by models such as the CNCPS.
This study was conducted to evaluate the effects of flint corn processing method (CPM) and level of NDF from roughage (rNDF) on performance, carcass characteristics, and starch utilization by finishing Nellore-based cattle fed high-concentrate, flint corn-based diets. In this study, 112 Nellore type bulls (initial BW 384.07 ± 29.53 kg and 24-36 mo of age) were individually fed using Calan gates or individual pens. The animals were used in a randomized complete block design in a 2 × 4 factorial arrangement with 2 CPM, high-moisture flint corn (HMC) or finely ground dry flint corn (FGC), with 1 of 4 levels of rNDF, 3, 8, 13, and 18% (DM basis), using sugarcane silage (SS) as roughage. Bulls were adapted to the finishing diet over a 21-d period and fed for a total of 81 d. Fecal starch (FS) concentration was determined on d 46 and 74 of the feeding period. There was a quadratic effect of rNDF on final BW (P < 0.01) and ADG (P = 0.01). Optimal concentrations of rNDF were estimated using the first derivative of second order polynomials, indicating that final BW and ADG were maximized with 13.3 and 13.0% rNDF, respectively. An interaction was observed between CPM and rNDF (P = 0.05) for DMI, with peak DMI occurring at 11.3 and 13.7% rNDF with FGC and HMC, respectively. Cattle fed HMC had 13.9% greater G:F (P < 0.01) compared with those fed FGC (0.172 vs. 0.151, respectively). There were quadratic effects of rNDF on HCW (P = 0.04) and ME intake (P < 0.01); heaviest carcass weights were estimated, in both cases, to be achieved with 12.8% rNDF. A quadratic effect of rNDF for renal, pelvic, and inguinal fat weight (P = 0.04) was observed, with a peak estimated to occur at 12.6% rNDF. An interaction between CPM and rNDF also was observed for FS (P < 0.05). Bulls fed FGC with 3% rNDF had greater FS content, and FS linearly decreased as concentration of rNDF increased. For bulls fed HMC, FS was 3.0% of DM and was unaffected by rNDF in the diet. Lower FS from bulls fed HMC suggests that availability of starch from flint corn was greater than that of FGC. For Nellore-based cattle fed a flint corn-based diet containing SS and 8% whole lint cottonseed, performance was optimized with 12.8% rNDF. In the absence of cottonseed addition to diets, optimal performance would be expected with about 14.5% rNDF.
BackgroundDespite the health concerns and nutritional importance of fatty acids, there is a relative paucity of studies in the literature that report genetic or genomic parameters, especially in the case of sheep populations. To investigate the genetic architecture of fatty acid composition of sheep, we conducted genome-wide association studies (GWAS) and estimated genomic heritabilities for fatty acid profile in Longissimus dorsi muscle of 216 male sheep.ResultsGenomic heritability estimates for fatty acid content ranged from 0.25 to 0.46, indicating that substantial genetic variation exists for the evaluated traits. Therefore, it is possible to alter fatty acid profiles through selection. Twenty-seven genomic regions of 10 adjacent SNPs associated with fatty acids composition were identified on chromosomes 1, 2, 3, 5, 8, 12, 14, 15, 16, 17, and 18, each explaining ≥0.30% of the additive genetic variance. Twenty-three genes supporting the understanding of genetic mechanisms of fat composition in sheep were identified in these regions, such as DGAT2, TRHDE, TPH2, ME1, C6, C7, UBE3D, PARP14, and MRPS30.ConclusionsEstimates of genomic heritabilities and elucidating important genomic regions can contribute to a better understanding of the genetic control of fatty acid deposition and improve the selection strategies to enhance meat quality and health attributes.
-The objective of this study was to evaluate the effect of addition of different levels of cottonseed (0, 14.35, 27.51 and 34.09 kg/100 kg) in the diet of Nellore steers on the meat characteristics. Thirty-six steers (average initial body weight of 333.50 and average age of 20 months) were kept in feedlot stalls for 94 days, each stall with three animals. The experiment was a completely randomized design with four treatments and nine replications. The values of protein, fixed mineral residue, shear force, lipid oxidation, meat color and fat color were similar among the treatments. The increase of the cottonseed levels in the diet reduced the amount of intramuscular fat and increased the moisture content in the meat linearly. Meat aroma and flavor were negatively influenced when cottonseed addition was greater than 27.51 and 34.09 kg/100 kg of the diet, respectively. Cottonseed diet did not modify the concentration of conjugated linoleic acid, saturated fatty acids or total unsaturated fatty acids, but linearly reduced the concentration of monounsaturated fatty acids in the meat.
