Considerable success has been obtained in the use of classical breeding to control economically important plant diseases, such as the coffee leaf rust and the coffee berry disease (CBD). There is a strong consensus that growing genetically resistant varieties is the most appropriate cost effective means of managing plant diseases and is one of the key components of crop improvement. It has also been recognized that a better knowledge of both, the pathogens and the plant defence mechanisms will allow the development of novel approaches to enhance the durability of resistance. After a brief description of concepts in the field of plant disease resistance, we attempt to give a view of the research progress on coffee leaf rust and CBD concerned with the pathogens infection and variability, coffee breeding for resistance and coffee resistance mechanisms. Há um grande consenso de que o uso de plantas geneticamente resistentes é o meio mais apropriado e eficaz em termos de custos do controle das doenças das plantas, sendo também um dos elementos chave do melhoramento da produção agrícola. Tem sido também reconhecido que um melhor conhecimento do agente patogênico e dos mecanismos de defesa das plantas permitirá o desenvolvimento de novas abordagens no sentido de aumentar a durabilidade da resistência. Após uma breve descrição de conceitos na área da resistência das plantas às doenças, nesta revisão tentou-se dar uma idéia do progresso na investigação da ferrugem alaranjada do cafeeiro e do CBD relativamente ao processo de infecção e variabilidade dos agentes patogênicos, melhoramento do cafeeiro para a resistência e mecanismos de resistência do cafeeiro. The hypersensitive response (HR) is a programmed cell death that is commonly associated with plant disease resistance. A novel lesion mimic mutant, vad1 (for vascular associated death1), that exhibits light conditional appearance of propagative HR-like lesions along the vascular system was identified. Lesion formation is associated with expression of defense genes, production of high levels of salicylic acid (SA), and increased resistance to virulent and avirulent strains of Pseudomonas syringae pv tomato. Analyses of the progeny from crosses between vad1 plants and either nahG transgenic plants, sid1, nonexpressor of PR1 (npr1), enhanced disease susceptibility1 (eds1), or non-race specific disease resistance1 (ndr1) mutants, revealed the vad1 cell death phenotype to be dependent on SA biosynthesis but NPR1 independent; in addition, both EDS1 and NDR1 are necessary for the proper timing and amplification of cell death as well as for increased resistance to Pseudomonas strains. VAD1 encodes a novel putative membrane-associated protein containing a GRAM domain, a lipid or protein binding signaling domain, and is expressed in response to pathogen infection at the vicinity of the hypersensitive lesions. VAD1 might thus represent a new potential function in cell death control associated with cells in the vicinity of vascular bundles.
Ultrastructural studies of 2,4-D (2,4 dichlorophenoxyacetic acid) induced coconut calli and of untreated controls enabled us to characterize early events in cellular reorganization leading to embryogenic cell individualization and subsequent development into proembryos. Embryogenic cells were characterized by special features that chie¯y aected the nucleus, cytoplasm and cell wall: deep invaginations of the nuclear envelope, proliferation of dictyosomes, with emission of Golgi vesicles, directly related to an increase in cell wall thickness. Modi®cation of the cell wall structure was studied and particular attention was paid to the cytolocalization of b-1,4-glucans, and of callose and pectin epitopes, using gold-conjugated probes. The ®rst changes (detected 7±14 d after 2,4-D increase) involved the closure of plasmodesmata, breaking of symplastic continuity, and callose deposition. The acquisition of embryogenic competence was linked to the appearance of an outer layer of ®brillar material containing pectin epitope (mainly un-methyl-esteri®ed), fully coating the embryogenic cells (21 d after the induction treatment). Some of the ultrastructural changes observed during the reprogramming of somatic cells towards embryogenesis can be likened to those accompanying the maturation of female gamete cells in many plant species. The possible signi®cance of these observations is discussed.
