1,25-Dihydroxyvitamin D3-induced differentiation in a human promyelocytic leukemia cell line (HL-60): receptor-mediated maturation to macrophage-like cells.

Abstract: The human-derived promyelocytic leukemia cell line, HL-60, is known to differentiate into mature myeloid cells in the presence of 1,25-dihydroxyvitamin D3 (1,25[OH]2D3) . We investigated differentiation by monitoring 1,25(OH)2 D3-exposed HL-60 cells for phagocytic activity, ability to reduce nitroblue tetrazolium, binding of the chemotaxin N-formyl-methionylleucyl-[ 3H]phenylalanine, development of nonspecific acid esterase activity, and morphological maturation of Wright-Giemsa-stained cells. 1,25(OH)2D3 conc… Show more

“…Treatment of leukemic cell lines with physiologic concentrations of 1,25(OH) 2 D induces their differentiation into monocytes; an initial proliferation is followed by growth arrest and terminal differentiation [23][24][25]. Recent studies show that 1,25(OH) 2 D binding to its receptor initiates a complex cascade of intracellular molecular events [26].…”

“…Phorbol esters induce the superoxide burst of HL-60 cells differentiated along the neutrophil or macrophage lineage with RA or calcitriol, respectively [14,17]. In vivo, the superoxide burst results in intracellular release of active oxygen species normally involved in destruction of phagocytosed microorganisms.…”

“…An alternative approach to the cytodestruction initiators of the differentiation of HL-60 cells, as well as of of cancer cells by conventional antineoplastic agents is other hematopoietic cell lines. 15,16 These effects are mediated through the induction of terminal differentiation, wherein cells by the binding of vitamin D 3 to a specific intracellular recepexposed to chemical or biological agents acquire the phenotor. 17 Clinical trials of vitamin D 3 in leukemic patients, howtypic characteristics of end-stage adult cell forms with no repever, have been ineffective, presumably due to hypercalcemia licative capacity and which ultimately undergo programmed which limited the therapeutic potential of this agent as a differcell death.…”
