1,25-Dihydroxyvitamin D3 upregulates the phosphatidylinositol signaling pathway in human keratinocytes by increasing phospholipase C levels.

Abstract: These studies indicate that 1,25(OH)2D3 modulates the keratinocyte signal transduction pathway by induction of phospholipase isoenzymes, a previously undescribed action for this hormone. (J. Clin. Invest. 1995. 96:602-609.)

“…In our prior studies using isolated colonic membranes, 1,25(OH) 2 D 3 failed to directly stimulate PI hydrolysis (4), making the involvement of these predominantly membrane-associated isoforms of PI-PLC in this phenomenon less likely. In human keratinocytes, however, 1,25(OH) 2 D 3 increased both the mRNA and protein abundance of PLC-␤ 1 (30). Given these latter observations, and the inherent complexities of studying regulated PLC activity in reconstituted in vitro experiments, this issue will require further investigation.…”

“…The addition of calcium leads to keratinocyte differentiation and the activation of several signaling pathways, one of which involves PLC-␥1, the predominant isoform of PLC in keratinocytes (13,14,16,17). However, it is not clear whether PLC-␥1 plays an essential role in the regulation of keratinocyte differentiation by calcium.…”

“…However, the keratin 1 gene is the only keratinocyte differentiation marker known to possess a 1,25(OH) 2 D 3 response element (5,6), and the expression of this marker is inhibited by 1,25(OH) 2 D 3 (7). In addition, 1,25(OH) 2 D 3 was recently shown to enhance the expression of several phosphoinositide-specific phospholipase C isoenzymes (8), the activity of which generates diacylglycerol (DAG). DAG, in turn, is known to regulate the activity of protein kinase C (PKC), and numerous data suggest the involvement of PKC in the regulation of keratinocyte growth and differentiation (reviewed in Refs.…”
