17β-Estradiol Potentiates the Stimulatory Effects of Progesterone on Cadherin-11 Expression in Cultured Human Endometrial Stromal Cells*

Abstract: Cadherin-11 (cad-11) is a novel member of the cadherin gene superfamily of calcium-dependent cell adhesion molecules. To date, the factors capable of regulating this cell adhesion molecule remain poorly characterized. We have recently determined that cad-11 expression in the human endometrium is tightly regulated during the menstrual cycle. The spatiotemporal expression of cad-11 in the stromal cells of the human endometrium during the menstrual cycle suggests that gonadal steroids regulate the expression of t… Show more

“…In general, the repertoire of cadherins present in eutopic endometrium and the levels of the mRNA transcripts encoding these CAMs were maintained in the endometriotic lesions. We and other authors have demonstrated that there is a direct correlation between cadherin mRNA and protein expression levels in a wide variety of human tissues and cells (Coutifaris et al, 1991;Chen et al, 1998;Getsios et al, 1998;Mialhe et al, 2000). Although we cannot determine whether there is a marked reduction in the expression of E-cad in the eutopic endometrial tissue samples obtained from these women, our studies suggest that significant levels of this CAM are expressed in the endometriotic lesions.…”

“…Enriched stromal cell cultures were isolated from endometrial tissues according to a previously described protocol [16]. Briefly, endometrial tissue samples were minced and subjected to 0.1% collagenase (type IV)/ hyaluronidase (type I-S, Sigma-Aldrich, St Lois, MO, USA) digestion in a shaking water bath at 37°C for 60 min.…”

“…The resultant cell pellet was washed once and resuspended in phenol red-free DMEM containing 25 mM glucose, L-glutamine, antibiotics (100 U/ml penicillin and 100 lg/ml streptomycin) and supplemented with 10% charcoal-stripped FBS. All the endometrial stromal cell cultures included in these studies were determined by immunocytochemical analysis, which was performed with a variety of markers, to have a purity of ≥99% [16,17].…”
