Abstract: Background: The hedgehog pathway plays a critical role in the development of prostate. However, the role of the hedgehog pathway in prostate cancer is not clear. Prostate cancer is the second most prevalent cause of cancer death in American men. Therefore, identification of novel therapeutic targets for prostate cancer has significant clinical implications.

“…Germline Sufu mutations have been identified in medulloblastoma (Taylor et al , 2002; Kool et al , 2014) and associated with the development of medulloblastoma in Gorlin syndrome (Pastorino et al , 2009; Kijima et al , 2012; Smith et al , 2014). Furthermore, somatic Sufu mutations have been identified in multiple other malignancies, including prostate cancer (Sheng et al , 2004). Sufu has a pivotal role in Hh signaling and is a relevant tumor suppressor, but the players involved in controlling Sufu levels are unknown and the relevance for cancer proliferation remains unrecognized.…”

“…In several cases, Smo-independent activation mechanisms have been documented, such as mutations in or loss of heterozygosity of the SUFU gene, which was found in medulloblastoma (20,21), prostate cancer (22), rhabdomyosarcoma (7), and a human lung cancer cell line (23). In addition, other downstream pathway activation mechanisms have been revealed, such as REN deletions (15), GLI gene amplifications (24)(25)(26), GLI1 chromosomal translocations (27), or GLI2 protein stabilization (28).…”

“…Recent studies have stressed the importance of Hh signaling in human prostate cancer (13)(14)(15)20). Elevated Hh signaling pathway activity may distinguish metastatic from localized prostate cancer, and pathway manipulation can modulate invasiveness and metastasis (13).…”
