1H NMR metabolic profiling of human serum associated with benign and malignant gallstone diseases

Abstract: Proton NMR based metabolic profile of serum associated with different gallbladder pathologies is presented. Quantitative and qualitative variations in the metabolic profile of serum in control samples and three different pathologies of gallbladder, chronic cholecystitis, xanthogranulomatous cholecystitis and carcinoma of gallbladder has been evaluated by use of 1 H NMR based metabonomics and multivariate chemometric methods. Multivariate partial least square discriminant analysis of 1 H NMR spectra showed a cl… Show more

“…Two-dimensional homo-nuclear ( 1 H- 1 H COSY and 1 H- 1 HTOCSY) and hetero-nuclear ( 1 H- 13 C HSQC) data were also acquired to confirm the resonance assignment. Assignment of metabolites was also confirmed from a standard database [21]. …”

“…NMR technology has been employed to analyze the lipid profiles of gallbladder tissue extracts, with the outcome that alterations in the lipid metabolism may be detected upon comparison between benign and malignant gallbladder diseases [222]. Additional research based on NMR analysis reported on the variations in the concentrations of low-density lipids (LDLs), very-low-density lipids (VLDLs), branchedchain amino acids (BCAAs), alanine, glutamine, creatinine, and tyrosine, 1,2-propanediol, pyruvate, glutamate, and formate in gallbladder cancer as compared to healthy individuals [223]. Moreover, gallbladder stones from patients from different geographical regions have been analyzed using solid-state NMR, showing different levels of cholesterol, bilirubin, and calcium carbonate correlations [224].…”

“…Biofluids, such as blood plasma, serum, urine, saliva, cerebrospinal fluid, prostatic secretions, seminal fluid, follicular fluid, bronchoalveolar lavage fluid (BALF), exhaled breath condensate (EBC), bile, faecal extracts and tissue/cell extracts, of cancer biopsy samples, as well as cancer cell lines, have been subjected to study by NMR for metabolite profiling . For NMR experiments, minimal sample preparation is required.…”
