“…Moreover, gene expression was negligible in the liver, kidneys, spleen, diaphragm, heart, lung and plasma following the intragastric, intraperitoneal and intravenous administration of pDNA (data not shown). These results are consistent with the results in our previous reports [23,24]. In intragastric and intravenous administration, naked pDNA would be degraded by digestive fluid in the stomach and nuclease in the blood.…”

“…Furthermore, we reported on site-selective gene expression following instillation of naked pDNA onto the liver surface [23][24][25][26], kidney surface [27] and gastric serosal surface [28] in mice. However, 30 µL of pDNA solution as an instillation volume was too large against the target organ in our previous studies; a large volume causes diffusion of pDNA solution to peripheral tissues.…”

“…In this regard, spleen surface instillation of naked pDNA is not stressful to the spleen, and thus repeated administration of naked pDNA might be possible by use of a catheter, as shown in our previous report on liver surface instillation of naked pDNA. 34) On the other hand, immunomodulatory cytokines could activate the immune system. There have been several investigations showing that co-delivery of antigen encoding genes and cytokine genes as vaccine adjuvants enhanced the immune response to DNA vaccination.…”

“…Furthermore, we reported on site-selective gene expression following the instillation of naked pDNA onto the liver surface [33][34][35][36] and kidney surface 37) in mice. The present study was undertaken to examine spleen-selective gene transfer following the administration of naked pDNA onto the spleen surface in mice.…”

The present study was undertaken to examine unilateral lung-selective gene transfer following the administration of naked pDNA onto the pulmonary pleural surface in mice.…”
