24-h Intraocular pressure control with evening-dosed travoprost/timolol, compared with latanoprost/timolol, fixed combinations in exfoliative glaucoma

Abstract: Purpose To evaluate 24-h efficacy of travoprost/timolol fixed combination (TTFC) vs latanoprost/timolol fixed combination (LTFC) in exfoliative glaucoma (XFG). Design A prospective, single-masked, crossover, active-controlled, randomized 24-h comparison. Methods After up to a 6-week medicine-free period, XFG patients were randomized to either TTFC or LTFC for 3 months, dosed each evening, and then changed to the opposite treatment for another 3 months. At the end of the washout, and both treatment periods, a 2… Show more

“…18 Past clinical studies showed that TTFC is superior to LTFC or DTFC in lowering IOP. [19][20][21] In contrast, other studies reported the opposite. 22,23 The reason for this discrepancy is unclear but might be explained by the design of future studies.…”

“…Although the difference in measurement time (AM or PM) was not significant in mixed-model analysis, others reported that travoprost was significantly more effective for reducing 24-hour IOP than latanoprost in other types of glaucoma (exfoliative glaucoma) patients. 21,27 Moreover, whereas past clinical randomized studies suggested that both LTFC and TTFC provided greater IOP reduction at evening dosing than at morning dosing, 28,29 the effect of time of dosing is not fully understood in NTG patients. In this study, all patients were administered both medications in the evening.…”

“…Recently, some new studies 12,13,17,19 that compare the three PG-timolol FCs have become available, and their efficacy and tolerability are still worthy of further exploration. Therefore, an updated systematic review and meta-analysis was undertaken to assess the clinical efficacy and tolerability of the three PG-timolol FCs.…”

“…Three fixed-combination prostaglandin analog/b-blocker formulations are currently available and include latanoprost plus timolol (LTFC; Xalacom*), travoprost plus timolol (TTFC; DuoTravy), and bimatoprost plus timolol (BTFC; Ganfortz; Allergan, Irvine, CA, USA) 10 . Recently, many clinical trials have compared the efficacy and tolerability of the three PG-timolol FCs [11][12][13][14][15][16][17][18][19] ; however, the results have sometimes been conflicting.…”

“…Furthermore, another study showed that evening-dose travoprost/ timolol fixed combination may provide greater 24-hour IOP reduction compared with evening-dose latanoprost/timolol fixed combination in XFG. 18 Most recently, bimatoprost/timolol fixed combination was shown to control mean 24-hour IOP significantly better than latanoprost monotherapy. 19 A receptor specifically upregulated in XFS is adenosine A3 receptor (AA3R).…”
