4-[3,5-Bis(2-hydroxyphenyl)-1,2,4-triazol-1-yl]- benzoic Acid: A Novel Efficient and Selective Iron(III) Complexing Agent

Abstract: An exceptionally stable 1:2 complex [FeL(2)](3-) is formed from the ligand H(3)L and Fe(III). In contrast, the affinity of this ligand for other biometals is relatively small. These properties make H(3)L a highly promising candidate for medical applications (e.g. for the treatment of iron overload).

“…In this procedure, chelator is added to the blood through a vein or administered orally in order to remove toxic element. Deferasirox (4-[3,5-bis(2-hydroxyphenyl)-1,2,4-triazol-1-yl]-benzoic acid, or ICL670; Figure 1) was first reported in 1999 (Heinz et al, 1999). It is a tridentate chelator with high selectivity for Fe 3þ , and its NO 2 donation arises from one triazole nitrogen and two phenolate oxygen donors.…”

“…9 LIC values derived from magnetic susceptometry as milligrams of iron per gram of liver tissue (wet weight) were multiplied by a factor of 3.33 to provide values per gram of liver tissue dry weight (mg Fe/g dw; the conventional units used for LIC). 10 Another technique used is a T2* imaging of heart. Low T2* values {< 8 ms compared with normal (> 20 ms)} are related to risk of heart failure and death in ironoverloaded thalassemia patients.…”

“…In agreement with the negative charge, [FeL 2 ] 3À is a slightly stronger base. Thus L 3À acts as tridentate coordinating ligand, where binding of the metal center occurs through the two phenolate groups and one of the nitrogen atoms of the heterocycle (Heinz et al, 1999).…”
