4‐Hydroxynonenal: A membrane lipid oxidation product of medicinal interest

Abstract: A comprehensive focus on 4-hydroxynonenal (HNE) as candidate molecule in a variety of pathophysiological conditions occurring in humans is here provided. Despite an active, now well characterized, metabolism in most cells and tissues, HNE can be easily detected and quantified by means of several methods, although with different sensitivity. Measurements of HNE and/or stable metabolites in biological fluids are already applied as lipid peroxidation/oxidative stress markers in a huge number of human disease proc… Show more

“…There is growing evidence (Armstrong, 2002; Poli et al, 2008) that HNE, generated during the LPO process and here reported to be present in AKU chondroptotic cartilage (Fig. 2), is a key mediator of oxidative stress‐induced pathophysiological effects.…”

“…As hyperglycaemia and HNE can trigger multiple signalling pathways [22,[26][27][28][29], in order to confirm the direct effect of WNT activation on renal fibrogenesis in diabetic nephropathy, primary HRPTC were exposed to WNT3A-conditioned medium (WCM); plain L cell conditioned medium (LCM) was used as a control. As shown by western blot analysis, WCM increased cytosolic and nuclear β-catenin levels significantly compared with LCM ( Fig.…”

“…Compared with the STZ-induced diabetic rats, Akita mice showed upregulation of more WNT ligands. In parallel, it has been reported that Akita mice develop more severe diabetic nephropathy compared with STZ-treated animals [29]. However, it has been reported that overexpression of Wnt10b was accompanied by a reduction of plasma levels of triacylglycerol as well as improved glucose homeostasis [31].…”

“…Although being an amphiphilic compound, 4-HNE is more concentrated in membranes where it reacts with membrane-bound proteins and phospholipids, compromising their functions. 87 Elevated 4-HNE level has been observed in both cortical and hippocampal tissues early after TBI in animal models, 88,89 and reduced 4-HNE level has been found in parallel with neuroprotective effects in animals treated with antioxidants. 90 Although in TBI-related studies, 4-HNE levels were evaluated in brain tissue, studies examining this compound in CSF and plasma have been carried out for other neurologic disorders such as Alzheimer's disease, amyotrophic lateral sclerosis, and stroke.…”
