5.ALPHA.,8.ALPHA.-Epidioxy-22E-ergosta-6,9(11),22-trien-3.BETA.-ol from an Edible Mushroom Suppresses Growth of HL60 Leukemia and HT29 Colon Adenocarcinoma Cells

Abstract: Mushrooms are widely consumed not only as foods but also as alternative medicines. The fungus-specific constituents of mushrooms are thought to be effective against cancer and other lifestyle-related diseases. Mushroom polysaccharides belonging to the b-glucans possess the antitumor and immunomodulating actions in vivo, and some clinical trials have been carried out with these agents. 1,2) Steroids are abundantly present in mushrooms 3,4) and were reported to have diverse physiological functions, such as anti-… Show more

“…The aqueous phase was then extracted with 1-butanol (1-BuOH), and the EtOAc-(3.8%), 1-BuOH-(5.2%), and H 2 O-(9.9%) soluble fractions were obtained. The EtOAc-and 1-BuOH-soluble fractions showed cytotoxic effects (IC 50 values of 120 and 199 mg/ml, respectively), and the H 2 O-soluble fraction showed less potent effects (IC 50 value Ͼ200 mg/ml).Through bioassay-guided isolation, 11) sitosterol (1, 0.00025%), 5a,8a-epidioxy-22E-ergosta-6,22-dien-3b-ol (2, 0.0028%), 12) 5a,8a-epidioxy-22E-ergosta-6,9(11),22-trien3b-ol (3, 0.0045%), 12) 5a,6a-epoxy-5a-ergosta-7,22-dien3b-ol (4, 0.00012%), 9) and ergosterol (5, 0.00024%) were obtained from the EtOAc-soluble fraction by ordinary-and reversed-phase silica gel column chromatography, preparative TLC, and HPLC (Fig. 1).…”

“…As a fatty acid, oleic acid (1) was purified, as sterols ergosterol peroxide (2), 9,11-dehydroergosterol peroxide (3), and cerevisterol (4). Substances were identified by spectral analysis using HRMS and onedimensional/two-dimensional NMR techniques and comparison of the data to the literature [20,21]. The structures of the isolated substances can be found in Figure 4.…”

“…In addition to ergosterol, several other ergostane and cholestane compounds have been isolated (Huang et al, 2002;Ueno et al, 1999;Marcotullio et al, 2008). Particularly interesting is the presence in the methanolic extract of S. imbricatus of ergosterol peroxide (or 3--hydroxy-5 ,8 -epidioxyergosta-6,22-diene) that shows a plethora of biological activities, such as antileukemic and anticancer (Kobori et al, 2006), apoptotic-inducing (Takei et al, 2005), and anti-inflammatory (Kobori et al, 2007). It is interesting to note that processing and cooking practices determine a lowering of nutrients concentrations and of antioxidant activity (Barros et al, 2007c).…”
