5-Hydroxytryptamine decreases somatostatin receptors and somatostatin-responsive adenylyl cyclase in rat pancreatic acinar membranes

Abstract: Pretreatment of pancreatic acini with 5-hydroxytryptamine (5-HT) reduced the binding of the labeled somatostatin (SS) analogue '%Ty?-SMS to pancreatic acinar membranes. This effect was dependent of the dose of 5-HT used and length of pretreatment. This inhibitory effect of 5-HT was abolished when pancreatic acini were pretreati with 5-I-IT in the presence of the 5-HT,r receptor-antagonist 5-hydroxytryptophyl-5-hydroxytryptophan amide (5-I-IV-DP). Pretreatment of pancreatic acini with 5-HT reduced the inhibitio… Show more

“…However, upon addition of 2 µ m SST‐14, no effect could be observed on the uptake of the small neutral amino acid MeAIB, acidic or basic amino acids such as glutamate or lysine, bioactive peptides such as deltorphin II or Leu‐enkephalin, or a non‐metabolizable glucose analogue such as 3‐methyl‐ O ‐glucose (Table 1). SST‐14 also failed to affect the uptake of a biogenic amine such as 5‐hydroxytryptamine, whose binding to pancreatic acini had been reported (Munoz‐Acedo et al . 1997) to be reduced by octreotide.…”
