5-Hydroxytryptamine increases contractile force in porcine right atrium but not in left ventricle

Abstract: Positive chronotropic as well as inotropic effects of 5-hydroxytryptamine (5-HT) have been observed in pig atrial tissue, but no data are available about the direct effects of 5-HT on ventricular tissue. In the present study we investigated inotropic effects of 5-HT on atrial and ventricular trabeculae obtained from hearts of 3 months old pigs. The baseline isometric contractile force was significantly higher in ventricular (4.14 +/- 1.25 mN) than in atrial tissue (0.47 +/- 0.11 mN). A noradrenaline concentrat… Show more

“…Peripherally, 5-HT4 receptors affect motility of the gastrointestinal tract and mediate increases in the porcine and human atria1 rate and contractility (Table 5). Interestingly, 5-HT4 receptors seem to be absent on porcine Schoemaker et al, 1992), as well as human (Jahnel et al, 1992;Schoemaker et al, 1993) ventricles.…”

“…The pig is one of the few species to possess atrial 5-HT 4 receptors, and porcine atrium is considered a good model for human atrial 5-HT 4 receptors ; Medhurst and Kaumann 1993;Schoemaker et al 1992). Activation of Gprotein-coupled 5-HT 4 receptors starts a complex signalling cascade involving cAMP-mediated activation of protein kinase A (PKA) that ultimately leads to positive inotropic and lusitropic responses in left and right atrium Sanders and Kaumann 1992).…”

“…In contrast, the inotropic effect was not observed in porcine [9] and human ventricles [13]. The effects were more complex in the canine heart [16], where direct negative inotropic and indirect positive inotropic effects were seen.…”

“…The inotropic effect of 5-HT has been reported to be direct in cat atria [5,6], guinea pig atria [5,7], rat hearts [8], porcine atriä [9], and human atria [10][11][12][13]; whereas indirect effects were reported for rabbit hearts [5,14,15]. In contrast, the inotropic effect was not observed in porcine [9] and human ventricles [13].…”
