64CuCl2 PET Imaging of 4T1-Related Allograft of Triple-Negative Breast Cancer in Mice

Abstract: 64CuCl2 is an economic radiotracer for oncologic PET investigations. In the present study, we characterized the uptake of 64CuCl2 in vivo by µPET/CT in an allograft 4T1-related mouse model (BALB/c) of advanced breast cancer. 18F-FDG was used as a comparator. Twenty-two animals were imaged 7–9 days following 4T1-cell implantation inside mammary glands. Dynamic 64CuCl2 µPET/CT acquisition or iterative static images up to 8 h p.i. were performed. Animal biodistribution and tumor uptake were first evaluated in viv… Show more

“…The development of novel radio-probes could significantly enhance the value of PET/CT imaging in breast cancer. [31][32][33][34][35] Some novel positron-labeled targeted molecular probes for TNBC are already in development, [36][37][38] however positron probes targeting EphA2 in TNBC have not previously been reported.…”

“…This study differs from previous ones in a few key aspects. Firstly, human cell lines were used to induce the tumor models, enhancing clinical translatability compared to studies using murine cell lines such as 4T1 [58][59][60]. Secondly, the positive cell line used in this study (MDA-MB-231) expresses native levels of PD-L1 without artificial overexpression, unlike some previous studies that have used human PD-L1 gene-transfected A375 cells and human PD-L1gene transfected B16-F10 [61,62].…”

“…Established modalities that reveal and evaluate the occurrence of BCa within murine models include bioluminescence imaging (BLI), micro-ultrasound imaging (MUI), magnetic resonance imaging (MRI), and positron emission tomography (PET), as summarized in Table 1 . These modalities have provided a better understanding and visualization of cancer development [ 28 ], early detection [ 29 ], and therapeutic efficacy [ 30 , 31 , 32 ] in cancers such as breast [ 29 , 31 , 32 ], prostate [ 33 ], colon [ 28 ], and lung [ 30 ]. BLI relies on the biochemical generation of light for tumor assessment while PET utilizes radiotracers to investigate tumor burden while also providing information on the metabolic and biochemical functions.…”
