Abstract: The pharmacokinetic characteristics of desglycinamide-arginine vasopressin (DGAVP) with respect to its transport across the blood-brain barrier (BBB) were studied with the use of serial CSF sampling in an individual animal and the unit impulse response methodology. Transport rate is determined as BBB clearance, the volume of plasma per unit time cleared of the peptide by BBB transport, and the extent of transport as the percentage of the administered dose transported into the central nervous system. Plasma kin… Show more

“…Several in vitro, in vivo, and in situ techniques for quantifying the passage of substances have been described. [64] The usual indicator for passage is the brain uptake index (BUI). This compares the ratio of the concentration of test substance taken up by the brain and the amount injected luminally with the ratio of the concentration of a reference substance (e.g.…”

“…The first study showing the feasibility of time course studies in CSF focused on heptabarbital [ 48 ]. Next the technique was applied to study the CNS distribution of desglycinamide-arginine vaspressin (DGAVP) as prototype peptide [ 49 ]. By applying the “unit impulse response” technology, analyzing the concentration profiles following intravenous and intracerebral administration, the input profile in the CNS could be determined [ 50 ].…”

“…In order to describe the tightness status of the BBB in vivo or in vitro, next to transendothelial electrical resistance (TEER), determining the permeability of sucrose, Evans blue and FITC labeled dextrans (FD) became the methods of choice. At least since the late 1980s it has been known that commercially available FDs contain various amounts of free FITC [5] and consist of a wide range of polymers with different molecular weights. Commonly, total fluorescence values have been measured in permeation studies without considering the size of the different fluorescent fractions.…”
