A 12-cistron Escherichia coli operon (hyf) encoding a putative proton-translocating formate hydrogenlyase system

Abstract: The nucleotide sequence has been determined for a twelve-gene operon of Escherichia coli designated the hyf operon (hyfASCD€FGH/R-focB). The hyf operon is located a t 5508-56.0 min and encodes a putative nine-subunit hydrogenase complex (hydrogenase four or Hyf), a potential formate-and 6%-dependent transcriptional activator, HyfR (related to FhIA), and a possible formate transporter, FocB (related to FocA). Five of the nine Hyf-complex subunits are related to subunits of both the E. coli hydrogenase-3 complex… Show more

“…Therefore, the block in CO 2 reduction lies between CO 2 and methylene-H 4 SPT. Finally, acetate was not converted to CH 4 and CO 2 at detectable levels by cell suspensions of either wild-type or Ech1 grown on methanol. Thus, we measured the oxidation of CO to CO 2 and H 2 , which represents the oxidative half of acetoclastic methanogenesis.…”

“…Recently, a novel family of multisubunit membrane-bound [NiFe] hydrogenases, which are closely related to proton-pumping NADH:quinone oxidoreductases (complex I in electron transport chain of mitochondria), has been recognized (1,2). This family includes hydrogenases 3 and 4 from Escherichia coli (3,4), CO-induced hydrogenase from Rhodospirillum rubrum (5,6), Eha and Ehb from Methanothermobacter species (1), and Ech hydrogenase (Ech) from Methanosarcina barkeri Fusaro (7). Various members of this family have been proposed to function in energy-conserving electron transport, reverse electron transport, or both.…”

“…Other highly-repressed genes include hyfD (4.3-fold), which encodes a hydrogenase membrane subunit (Andrews et al, 1997), frlA (3.0-fold), which encodes a fructoselysine transporter (Wiame et al, 2002) and yieL (7.0-fold), which encodes a putative xylanase; all three loci are repressed in LB glu biofilms by the AI-2 exporter TqsA (Herzberg et al, 2006). Finding differential expression of these biofilm-related genes on deletion of yncC supports Genes studied by a gel shift experiment with YncC.…”
