A bacterial DNA quadruplex with exceptional K+ selectivity and unique structural polymorphism

Abstract: The G-quadruplex forming sequence d[(G4CT)3G4] shows complete and continuous quadruplex interconversion upon increasing K+-concentrations and pronounced K+ selectivity.

“…An involvement of quadruplex structures in antigenic variation was also suggested in Borrelia burgdorferi (Walia and Chaconas, 2013) and Treponema pallidum (Giacani et al, 2012). One of those motifs was recently characterized by our group and found to be significantly enriched in bacteria (Rehm et al, 2014). Apart from intergenically occurring quadruplexes, four-stranded motifs also occur in open reading frames (ORFs).…”

“…The distance distributions extracted between pairs of paramagnetic reporter molecules provide constraints to the range of possible conformations the macromolecule can adopt. DEER previously has been used to map conformations of human telomeric quadruplexes 5254 and a G-rich sequence found in Treponema pallidum 55 . These studies utilized site directed spin labeling, in which nitroxide spin labels are attached covalently to a deoxyribose group, to probe quadruplex conformation and folding motifs.…”
