A behavioral screen for isolating zebrafish mutants with visual system defects.

Abstract: Optokinetic and phototactic behaviors of zebrafish larvae were examined for their usefulness in screening for recessive defects in the visual system. The optokinetic response can be reliably and rapidly detected in 5-day larvae, whereas the phototactic response of larvae is variable and not robust enough to be useful for screening. We therefore measured optokinetic responses ofmutagenized larvae as a genetic screen for visual system defects. Third-generation larvae, representing 266 mutagenized genomes, were e… Show more

“…Given a choice between dark and light, both larvae and adults have a tendency to prefer the illuminated part of their tank (Brockerhoff et al, 1995). However, this preference is not robust enough to be practical for distinguishing visual system function between mutant and normal animals.…”

“…Its development closely follows the morphologic maturation of the retina. By 5 dpf, electroretinograms (ERG) containing all the components of a typical vertebrate ERG can be recorded (Branchek, 1984;Brockerhoff et al, 1995;Hughes et al, 1998;Saszik et al, 1999;Bilotta et al, 2001;Seeliger et al, 2002).…”
