A brief perspective on the diverging theories of lymphatic targeting with colloids

Abstract: For targeted delivery of colloids to the lymphatic system, the colloids should efficiently reach and remain in the lymphatics for a considerable period of time. As per the current knowledge, diffusion and phagocytosis are the two mechanisms through which colloids reach the lymphatic system. Several parameters including particle size and charge have been shown to affect the direct uptake of colloids by the lymphatic system. Although many researchers attached ligands on the surface of colloids to promote phagocy… Show more

“…On the other hand, CA as a lipid moiety, could facilitate intestinal absorption through lymphatic uptake as previously observed and reported by Garg et al, Makwana et al and Baek et al [67,68,69,70]. Moreover, Siram et al stated that nanovesicles and nanoparticles with a PS less than 100 nm could be capable of achieving better lymph uptake, which applies to the nanovesicles reconstituted from the optimized IFN formulations [71]. Enhancing the lymphatic uptake of RC could bypass the first pass effect and hence increase the oral bioavailability [72].…”

“…As the channels in cancerous lymph node walls can reach about 500nm, while only about 30-50nm in capillary walls [14]. Besides, the phagocytosis and pinocytosis effects of macrophages can also help the nanoparticles reach the lymphatic nodes [15]. Nanoscale carriers available for lymphatic targeting chemotherapy include liposomes, dendrimers, polymer micelles, carbon nanoparticles, etc [16].…”
