A Characterization of Estrogen’s Influence on Anterior Pituitary Androgen Receptor: Effect of Bromocriptine Treatment

Abstract: The action of gonadal steroids are mediated through the binding to specific intracellular receptors. Our recent studies have demonstrated that estrogen (E) treatment will increase the concentration of cytosolic androgen receptor (AR) in the anterior pituitary (AP) gland perhaps reflecting in increased sensitivity of this tissue circulating androgen. However, chronic E treatment also increases AP weight and cell number. Consequently, the following studies were performed to determine the relationship between inc… Show more

“…To clamp the estrous cycle changes of the steroid hormones (Cheung and Hammer, ; Sarkar and Minami, ), we employed the procedure of ovariectomy and implantation of an estrogen capsule to maintain the animals in an estrogenic phase that is known to maintain the activity of POMC neurons in an elevated phase (Bohler et al., ). The capsule containing estradiol has been reported to maintain plasma levels about 75 to 100 pg/ml, similar to those observed during the preovulatory phase of the estrous cycle (Handa and Rodriguez, ; Lino et al., ). Both male and female rats were killed at PD90 at 10 pm , the brains were dissected, MBH tissue samples were collected and immediately frozen for further analyses.…”

“…Although there is little precedent for cyclic changes in AR expression in neural tissue, AR expression in the ovary has been shown to fluctuate with the estrous cycle in rats (Szoltys and Slomczynska, 2000). Evidence of estrogenic up‐regulation of AR detected by binding assays and in situ hybridization has been demonstrated in some rat brain areas and pituitary (Handa et al, 1987, 1996; Handa and Rodriguez, 1991; McAbee and Doncarlos, 1999). Whether estrogens alter the level of AR mRNA in peripheral nerves from females requires further study.…”

“…Such work has been carried out over many years with each group focusing on one or a few genes. Categories of genes whose expression have been studied are those encoding hormones and hormone receptors [1][2][3][4][5], paracrine regulators of cell proliferation and their receptors [6][7][8][9][10][11][12], tumor suppressors [13,14], intracellular signaling proteins [5,15,16], proteases [5,17,18], and angiogenic factors and their receptors [9,19,20].…”
