A Chinese Herbal Formulation, Xiao-Er-An-Shen Decoction, Attenuates Tourette Syndrome, Possibly by Reversing Abnormal Changes in Neurotransmitter Levels and Enhancing Antioxidant Status in Mouse Brain

Abstract: Xiao-Er-An-Shen Decoction (XEASD) has been used clinically for the treatment of Tourette syndrome (TS) in children for more than 20 years in mainland China. The biochemical mechanism underlying the therapeutic action produced by XEASD treatment against TS remains unknown. However, a previous study has shown that pre-incubation of PC12 neuronal cells with XEASD can induce neurite outgrowth and protect against oxidative stress. In the present study, using a mouse model of TS induced by 3,3’-iminodipropionitrile … Show more

“…With excessive production of ROS and/or depletion of antioxidants, ROS balance will be disrupted and cause oxidative stress [37] . Interestingly, mitochondria are also the main targets of ROS damage, which in turn destroys the function of mitochondria and leads to excessive ROS generation [43][44][45] . Excessive oxidative stress can lead to the collapse of ΔΨm and mitochondrial membrane structure [46,47] .…”

“…IDPN is a neurotoxin that is commonly used to establish TS animal models. 30 In this study, stereotypy scoring, nodding experiment, and autonomic activity test were conducted to access the severity of behavioral symptoms of rats with TS induced by IDPN. The stereotypy score and nodding numbers can objectively reflect the severity of tics.…”

“…There has also been increasing modern medical research in China to understand the mechanisms of TCM. For example, another commonly used TCM decoction (Xiao-Er-An-Shen) for the treatment of TS in children in mainland China, its beneficial effect was shown to be associated with reversing abnormal changes of neurotransmitter levels and enhancing antioxidant status in an experimental mouse model of TS (119).…”
