A Concise Review on the Use of Mesenchymal Stem Cells in Cell Sheet-Based Tissue Engineering with Special Emphasis on Bone Tissue Regeneration

Abstract: The integration of stem cell technology and cell sheet engineering improved the potential use of cell sheet products in regenerative medicine. This review will discuss the use of mesenchymal stem cells (MSCs) in cell sheet-based tissue engineering. Besides their adhesiveness to plastic surfaces and their extensive differentiation potential in vitro, MSCs are easily accessible, expandable in vitro with acceptable genomic stability, and few ethical issues. With all these advantages, they are extremely well suite… Show more

“…The grafting of a tissue‐engineered construct seeded with adult mesenchymal stem cells (MSCs) into injured and ischemic tissues has emerged in the past decade as a potential therapeutic option for bone repair and regeneration . The success of such an approach is directly conditioned by the establishment of a fully functional vascular network within the grafted tissue construct to allow its survival.…”

“…The cell sheet technique is based on implantation of cells grown as single or multiple layers together with their secreted extracellular matrix (ECM), as opposed to conventional single‐cell suspensions. Proposed advantages of this technique include preservation of the cell‐to‐cell connections and ECM components along with a high cell‐seeding efficacy (Yorukoglu, Kiter, Akkaya, Satiroglu‐Tufan, & Tufan, ). Superior NBF after 4 months and comparable volumetric stability of augmented sites after 6–12 months were observed in the POC‐seeded AB (+PRP) grafts compared to conventional AB (+PRP) grafts (Nagata et al., ; Ogawa et al., ).…”

“…Examples on MSC-delivered genes are SDF-1 to ameliorate MI and ischemic brain injury (Penn and Khalil, 2008), glucagon-like peptide-1 to reduce amyloid deposition in Alzheimer's brains (Klinge et al, 2011), and IL-10 to restrain collagen-induced arthritis (Choi et al, 2008). Architectural MSC engineering involves cell culturing to obtain cellular sheets which can be further maintained in organspecific stimulating media or assembled onto organ scaffolds to restore injured or defective tissue [e.g., bone regeneration (Yorukoglu et al, 2017) and spinal cord injury (Zeng et al, 2011) applications].…”
